A wound closure device for use in sealing a tissue perforation created by an access device, comprising: an elongate body having a distal portion and a proximal portion, and a plurality of barbs extending from a surface thereof and extending at least partially in a distal direction; wherein the elongate body is a suture, a plug member having a tissue facing surface coupled to the distal end of the elongate body, wherein the elongate body, the plug member, or both, comprise at least one reactive group selected from the group consisting of isocyanate, N-hydroxy succinimide, cyanoacrylate, aldehyde, genipin, polylysine, diimide, diisocyanate, cyanamide, carbodiimide, dimethyl adipimidate, and combinations thereof; wherein the wound closure device further comprises an inner and a relatively rigid outer member each having an opening extending therethrough and each moveably positioned on the elongate body.
