Each chamber of the first and second multi-chamber devices holds a component of the sealing material, and operating the valve assembly permits mixing of at least some of the components prior to connecting the sealing material applicator to the delivery member. The expandable member is positionable within the vessel to temporarily seal closed the vessel puncture from within the vessel. The closure device is operable to deliver the sealing material from the sealing material applicator, through the delivery member, and to the tissue tract to seal closed the vessel puncture from outside the vessel.
