The invention provides a method of producing an in-focus image of an area on a sample plate for an ion source, e.g., a matrix-based ion source or any other type of ion source that employs a sample plate onto which samples are deposited. The method generally includes: a) positioning an area of the sample plate in the field of view of an imaging device; b) producing a plurality of images of the area having different in-focus regions; and c) generating an in-focus image of the area using the plurality of images. The in-focus image may be two-dimensional or three-dimensional. Systems and programming for performing the methods are also provided.

A camera having an automatic focus (AF) and an image-shake compensating device. The automatic focus device includes a phase-shift detector having photodetecting means (7,8,18) for receiving light passing through different portions (3,4) of the main imaging lens pupil and means for detecting the focus condition of the main imaging lens (10) from the light distributions received by the photodetecting means. The image-shake compensating device includes vibration detecting means (26) and actuating means (28) for either displacing a lens (10b) perpendicularly to the optical axis of the main imaging lens (10) or pivoting the whole lens system about an axis perpendicular to the optical axis. The camera includes also means (27) for detecting the de-alignment of the lens system caused by the actuating means, memory means (22) for storing aberration information in dependence of the lens system de-alignment and means (19,21) for calculating the amount of movement of the focusing lens along the optical axis on the basis of the detected focus condition and aberration information read out from the memory means. The system is capable of achieving proper focusing by correcting the focusing position taking into account the different spectral sensitivity of photographic material and photodetectors (7,8,18) even when additional chromatic or spherical aberrations are produced by the image-shake compensating device.
