The invention relates to a new dipstick assay for detecting and quantifying the content of an target analyte in a sample. The assay is particularly useful for example in the diagnosis and monitoring of alcoholism by the detection of asialo transferrin or carbohydrate free transferrin (CFT). Thus, provided is a dipstick for determining the content of a target analyte variant in a mixture of analyte variants in a sample, comprising: a) a sample application zone, b) a screening zone having an immobilised binding ligand having a binding affinity for a non-target analyte variant or varinats, c) a conjugate zone comprising a detector reagent, d) a reading zone for detection of said analyte.

The invention provides a method of assessment of carbohydrate-deficient transferrin in a transferrin containing body fluid, said method comprising the steps of:i) obtaining a transferrin containing liquid sample of or derived from a said fluid;ii) contacting said sample with a source of iron ions;iii) subsequently contacting said sample with an anionic ion exchange resin at a pH such as to cause carbohydrate-deficient transferrin to be retained by said resin;iv) subsequently contacting said resin with an eluant serving to release carbohydrate-deficient transferrin into the eluate from said resin;v) collecting a volume of said eluate substantially free from tetra- and penta-sialo transferrin; andvi) assessing the transferrin variant content in said volume of eluate.By including at least a proportion of the trisialotransferrin in the eluate, it is possible to use relatively simple assessment techniques such as turbidimetry in the assay.

Colorimetric indicators for breath, air, gas, & vapor analyses that can be thrown away after a single use, are easy to use, low cost, small, and simple to manufacture for indicating at least volume passed through the colorimetric indicators and may also be used to indicate concentration of an analyte within breath, air, gas, & vapor passed through the colorimetric indicators. The present invention is directed to colorimetric indicators for breath, air, gas, & vapor analyses. The colorimetric indicators may have a housing with an entrance port, an outlet, a volume indicator, and an analyte indicating reagent. The volume indicator may be a breath volume reagent. Different analytes may be measured, the analyte indicating reagent depending upon the particular analyte being to monitored. The breath volume indicator is colorimetric, and changes color when a predetermined volume of air is passed through the colorimetric indicators for breath, air, gas, & vapor analyses. The analyte indicator reagent is also colorimetric, and changes color when a predetermined amount of chemical concentration of the analyte is exposed to the analyte indicator reagent. A process for manufacturing a colorimetric indicator for breath, air, gas, and vapor analyses, comprises at least the following steps: immobilizing a reagent, such color change inducing reagent being composed of at least one constant concentration component.

An antibody is produced to an antigen, the antibody being cross-reactive with acetaldehyde-protein condensates, these condensates including protein moieties corresponding to or different from the protein moiety of the antigen. Detection and measurement of acetaldehyde-protein condensates may be performed by reacting the antibody with the acetaldehyde-protein condensate to form a complex and measuring the complex.
