摘要: Embodiments of the disclosed subject matter provide an automated method and system to isolate and collect cells using computerized analysis of images of cells and their surroundings obtained from a digital imaging device or system. Embodiments of the disclosed subject matter make use of a “microwell array,” which can comprise a formed, elastomeric grid of indentations or “wells.” Many, most, or all of the wells in a microwell array can contain a releasable, microfabricated element, which can be referred to as a “raft.” Embodiments of the disclosed subject matter provide a system and method for cell collection that includes computerized identification and collection of rafts with isolated single cells or a specific group or groups of cells, eliminating the need for continuous human identification and selection.

摘要: Described herein are binding proteins useful against ACE2-targeted viruses (e.g., SARS-CoV and SARS-CoV-2, etc.), and methods of using them. These binding proteins may include an extracellular portion of angiotensin-converting enzyme 2 (ACE2), excluding the collectrin domain, and a flexible polypeptide flexible linker coupling the ACE2 portion to a fragment crystallization (Fc) domain. These binding proteins dimerize, and the flexible linker may be chosen to be sufficiently long to permit concurrent interaction with multiple Spike (S) proteins on the ACE2-targeted virus.

摘要: Viral promoters and compositions and methods of use thereof are provided. Compositions include viruses with impaired ability to reactivate from latency, and pharmaceutical compositions and method of use thereof. The genome of the viruses include one or more mutations that reduce expression from one or more promoters that regulate expression of viral genes during reactivation from latency. The mutation(s) are typically in a region of the viral genome that includes (i) promoter elements of the iP1 promoter of human cytomegalovirus, or the sequence of another virus corresponding thereto (e.g., an iP1 promoter homolog); (ii) promoter elements of the iP2 promoter of human cytomegalovirus, or sequence of another virus corresponding thereto (e.g., an iP2 promoter homolog); or (iii) a combination thereof. In some embodiments the virus encodes one or more heterologous antigens. The viruses can be used as vaccines to induce prophylactic and therapeutic immune responses in subjects in need thereof.

摘要: Systems and methods for measuring micro-architectural properties of vascular networks are described herein. An example method can include injecting an ultrasound contrast agent into a medium, and transmitting a plurality of acoustic pulses into the medium using a ultrasound transducer array. Each respective acoustic pulse can be transmitted from one or more elements of the ultrasound transducer array. The method can also include receiving a plurality of backscattered signals with the ultrasound transducer array in response to each respective acoustic pulse, and obtaining a response matrix including the backscattered signals. The method can further include extracting a coherent or incoherent contribution to the backscattered signals from the response matrix, and quantifying a property of a vascular network based on the coherent or incoherent contribution to the backscattered signals.

摘要: Disclosed are antimicrobial compositions and formulations directed against a broad range of bacteria and Pseudomonas aeruginosa biofilms. In an aspect, the compositions and formulations do not engender resistance. In an aspect, in combination with antibiotics, the compositions and formulations slow the development of antibiotic-resistance and greatly improve bacterial susceptibility to multiple classes of antibiotics.

摘要: This specification describes systems and methods for using Zero Echo Time (ZTE) magnetic resonance imaging (MRI) sequences for applications to functional MRI (fMRI). In some examples, a system for functional magnetic resonance imaging includes a magnetic resonance imaging (MRI) scanner and a control console implemented on at least one processor. The control console is configured for executing, using the MRI scanner, a zero echo time (ZTE) pulse sequence; acquiring, using the MRI scanner, magnetic resonance data in response to the ZTE pulse sequence; and constructing at least one MRI image using the magnetic resonance data and measuring tissue oxygenation (PtO2)-related T1 changes as a proxy of neural activity changes of a subject using the at least one MRI image.
