摘要: A cardiovascular graft is provided, such as a prosthetic heart valve or blood vessel, that comprises mesenchymal stem cells (MSCs) and/or progeny thereof, modified to knock down or knockout expression of the telomerases reverse transcriptase gene (TERT), or otherwise reduce activity of TERT. Also provided are a method of making the cardiovascular graft, and a method of implanting the cardiovascular graft.

The methods include administering to a subject a therapeutically effective amount of a dominant negative tumor necrosis factor (DN-TNF)-α protein and/or a nucleic acid encoding the DN-TNF-α protein. 摘要: This document provides methods and materials for expanding tumor infiltrating γδ T cells (e.g., tumor infiltrating γδ T cells) in culture. For example, methods and materials for expanding large numbers of tumor infiltrating γδ T cells (e.g., tumor infiltrating γδ T cells that are predominantly Vδ1+) from tissue obtained from a mammal having cancer (e.g., a tumor sample), an autoimmune condition, or an infection are provided. Populations of such tumor infiltrating γδ T cells and methods and materials for using such tumor infiltrating γδ T cells and/or such populations to treat cancer within a mammal (e.g., a human) also are provided.

摘要: Provided herein are methods of identifying a protein capable of binding a ligand, the method comprising: (a) contacting the ligand with two or more samples comprising a plurality of proteins in a solution; (b) separating the proteins bound to the ligand (“bound proteins”) from the proteins that are not bound to the ligand (“unbound proteins”) in each sample; (c) denaturing and digesting the bound proteins to form a plurality of peptides in each sample; (d) quantifying a plurality of molecular features contained in the plurality of peptides in each sample, wherein the molecular features are defined as having a mass to charge ratio, retention time, and peak intensity as measured by mass spectrometry; and (e) ranking the molecular features that exhibit a statistically significant difference in quantity between the samples contacted with the ligand and a sample that is not contacted with the ligand (“statistically significant molecular feature”).

摘要: The present invention relates to oncolytic vaccinia viruses which have been modified to promote anti-tumor immunity and/or reduce host immunity and/or antibody response against the virus. It is based, at least in part, on the discovery that oncolytic vaccinia virus (i) bearing a genome deletion of a gene that reduces T cell immunity (interleukin-18 binding protein); (ii) treated with a sialidase enzyme which is believed to reduce TLR2 activation and therefore the antibody response; (iii) carrying a gene that enhances cytotoxic T lymphocyte induction (e.g., TRIF) and/or (iv) reduces tumor myeloid-derived suppressor cells by reducing prostaglandin E2 reduces tumor growth. Accordingly, the present invention provides for immunooncolytic vaccinia viruses and methods of using them in the treatment of cancers.
