    摘要: The invention provides methods for treating or preventing bone degenerative disorders. The disorders treated or prevented include, for example, osteopenia, osteomalacia, osteoporosis, osteomyeloma, osteodystrophy, Paget's disease, osteogenesis imprerfecta, and bone degenerative disorders associated with chronic renal disease, hyperparathyroidism, high levels of endogenous thyrotropin, and long-term use of corticosteroids. The disclosed therapeutic methods include administering to a mammal a TSHR agonist in an amount effective to: (1) treat or prevent a bone degenerative disorder; (2) slow bone deterioration; (3) restore lost bone; (4) stimulate new bone formation; and/or (5) maintain bone mass and/or bone quality. TSHR agonists such as thyrotropin and its modified forms are provided along with other compounds, such as anti-resorptive agent and bone metabolic agents.

    摘要: Disclosed herein are methods and compositions for identifying morphogen analogs. The preferred methods and compositions relate to the discovery that morphogen upregulation of the mouse type X collagen promoter activity is mediated by a MEF-2 like sequence and requires an adjacent AP-1 sequence. Certain methods rest on the use of test cells comprising DNA defining a morphogen-responsive transcription activating element operatively associated with a reporter gene. Other methods rest on the use of DNAs for measuring morphogen-inducible DNA-binding. In certain preferred embodiments, the methods and DNAs involve an osteogenic protein 1 (OP-1) responsive transcription activating element. Substances that mediate interaction with and/or activate the OP-1 responsive transcription activating element are considered herein likely to be useful for reproducing in vivo effects of morphogens such as OP-1.

    摘要: Disclosed are methods and compositions for maintaining neural pathways in a mammal including: enhancing survival of neurons at risk of dying; inducing cellular repair of damaged neurons and neural pathways; stimulating neurons to maintain their differentiated phenotype; and promoting dendritic outgrowth, including maintaining dendritic arbors and regenerating dendritic architecture. In one embodiment, the invention provides means for stimulating CAM expression in neurons. The invention also provides means for evaluating the status of nerve tissue, including means for detecting and monitoring neuropathies in a mammal. The methods, devices and compositions include a morphogen or morphogen-stimulating agent provided to the mammal in a therapeutically effective concentration. In another embodiment, the invention provides methods and compositions which include a morphogen or morphogen-stimulating agent, and a nerve trophic factor or nerve trophic factor-stimulating agent at concentrations effective for stimulating dendrite outgrowth. The morphogen and the nerve trophic factor can be admixed in combination.
