One of the supply reservoir element and the microfluidic device includes a puncturing element in order to cause fluid flow of the liquid reagent from the first reservoir unit to the reception device when the puncturing element is operated. A system and method for providing an improved microchip measurement apparatus comprising an external reservoir connected via an input line and output line to a well disposed on the microchip. The reservoir and well may be sealed and a pump connected to the input line so as to enable a continuous flow of liquid from the reservoir to the well. In addition, the volume of liquid in the well may be kept constant, thereby improving the overall measurement accuracy.

A transgenic mouse with alterations in a PA28&bgr; gene is prepared by introduction of an altered PA28&bgr; gene into a host mouse. The resulting transgenic animals do not produce functional PA28 molecules. Cells and cell lines derived from these animals also contain the altered PA28&bgr; gene.
