The improved techniques can pertain to creating, publishing, hosting, accessing, subscribing, managing, transferring, and/or playing podcasts. According to one aspect, a client application can subscribe to podcasts and then automatically monitor the podcasts for updates to be downloaded. In the event that user interest in a podcast becomes inadequate, downloading of further updates can be restricted. According to another aspect, a podcast can be subscribed to through use of a portable subscription file. According to still another aspect, podcast feeds can be enhanced to include segment elements and other metadata.

Such tags can include a block tag that may be embedded into a syndication feed to indicate whether the feed is to be blocked from being included in the online media store. According to another aspect, management of podcasts may be facilitated by other mechanisms. In accordance with one embodiment, a user may challenge the ownership of a podcast to block or remove a podcast from the online media store. In accordance with another embodiment, images may be stored to allow efficient retrieval of the corresponding episode(s) of a podcast.

    摘要: The disclosed embodiments relate generally to the downloading of media items. The media items may include, for example, audio, video, image, or podcast data. In accordance with one embodiment, two or more media items may be concurrently downloaded. In accordance with another embodiment, a user may control the downloading of each of the media items, as desired. More particularly, the user may choose to download, pause, or resume downloading one or more media items identified in the list of media items. In other words, the user may alter the download status of any of the items in the list of media items. In addition, the user may re-prioritize the order in which the media items in the list or a subset thereof are to be downloaded.

    摘要: The present invention provides novel engineered derivatives of green fluorescent protein (GFP) which have an amino acid sequence which is modified by amino acid substitution compared with the amino acid sequence of wild type Green Fluorescent Protein. The modified GFPs exhibit enhanced fluorescence relative to wtGFP when expressed in non-homologous cells at temperatures above 30° C., and when excited at about 490 nm compared to the parent proteins, i.e. wtGFP. An example of a preferred protein is F64L-S175G-E222G-GFP. The modified GFPs provide a means for detecting GFP reporters in mammalian cells at lower levels of expression and/or increased sensitivity relative to wtGFP. This greatly improves the usefulness of fluorescent proteins in studying cellular functions in living cells.

    摘要: The present invention, generally speaking, provides a powerful yet easy to use electronic publishing tool for information providers and online service operators. The electronic publishing tool allows information providers to manage the content they provide to online services. It also reduces the effort required by operations staff of online services to support a growing number of information providers. Finally, the electronic publishing tool allows content to be captured from information providers (and, by extension, subscribers) so that it can be published onto a variety of platforms with minimal additional effort. For example information, in addition to or in lieu of being published on an online service, may be published on CD through a facility such as AppleLink CD (AppleLink CD allows a portion of the "static" content of an online service such as AppleLink to be published and distributed in CD form, with the result that the content is infinitely reusable without incurring connect charges.) Furthermore, information previously published on one online service may be published on one or more other online services, or information may be published on several online services at the same time. A number of potentially "thorny" problems of distributed database systems are solved in an elegant and efficient manner. The first issue involves synchronizing different copies of an item on machines that are not continuously linked at which each copy may be changed independently. Rather than attempting to formulate a set of synchronization policies generally applicable to all items in all instances, synchronization policies are moved from the database engine to the items themselves. The second issue involves lifetime--knowing, given a complex set of interrelationship of items, when an item may be safely deleted. This problem is addressed by providing an interested party mechanism whereby items may "express interest" in each other. When no item is interested in a particular item, that item may safely "go away."
