    摘要: The invention provides methods, compositions, systems, and kits that include an enzyme/substrate co-delivery system. The liquid delivery system includes at least one enzyme encapsulated in a water-soluble polymeric matrix and a substrate for the enzyme in a carrier liquid in which the polymeric matrix is insoluble. When water is added, the polymeric matrix is solubilized and enzyme is released from the matrix, permitting catalytic action upon the substrate.

    摘要: A fluidized bed coating apparatus is provided and includes a fluid inlet adapted to be connected to a source of fluidizing fluid. The fluid inlet has an entrance opening having a first diameter and an exit opening downstream of the entrance opening. The exit opening has a second diameter that is larger than the first diameter. The inlet also includes an expanding inlet section, between the entrance opening and the exit opening, that has an average diameter that increases in a direction from said entrance opening to said exit opening. A fluid distribution plate is provided adjacent the exit opening and includes a plurality of through-passages to allow the even and distributed passage of a fluidizing fluid therethrough. A chamber having a chamber inlet and a chamber outlet is provided and the fluid distribution plate is positioned between the exit opening and the chamber inlet. The chamber includes at least a first diverging section having an average diameter that increases in the direction from the chamber inlet downstream toward the chamber outlet. The symmetrical design of the coater provides an evenly distributed flow of fluid that enables efficient and complete fluidization and coating of very large particle beds.

    摘要: A method is provided for the extraction of water soluble biomaterials such as enzymes or proteins into carbon dioxide utilizing certain carbon dioxide soluble surfactants. The extraction can be performed on an aqueous solution, a fermentation broth or a fluid. The method includes the process steps of forming a carbon dioxide/surfactant mixture which involves dissolving carbon dioxide soluble surfactant(s) in carbon dioxide. The carbon dioxide can be in a liquid or supercritical form and the surfactant includes tail and head groups that interact with the biomaterials. Further, the mixture is added to the aqueous solution, fermentation broth or liquid under conditions to allow for extraction of the biomaterials. The method further includes depressurizing and/or temperature adjusting to remove the water soluble biomaterials. The surfactants include fluroethers, oligomers of propylene-oxide, siloxanes, etc. The biomaterials include proteins or enzymes. The carbon dioxide is suberitical or supercritical.

    摘要: Granules are prepared containing an admixture of protein and starch layered over an inert particle. Proteins include pharmaceutically important proteins such as hormones, or industrially important proteins such as enzymes including proteases, amylases, lipases and cellulases capable of hydrolyzing substrates such as stains. Inert particles include inorganic salts, sugars, sugar alcohols, small organic molecules such as organic acids or salts, and minerals such as clays or silicates. The admixture may also contain sugar such as sucrose. A ratio of corn starch to sugar much greater than 1:1 such as in a range of about 5:1 to about 15:1 is preferred. A coating layer may be between the inert particle and the admixture and/or over the admixture. Methods that may be used in preparing the granules include pan-coating, fluid-bed coating, prilling, disc granulation, spray drying, extrusion, centrifugal extrusion, spheronization, drum granulation and high shear agglomeration.
