摘要: Provided is a cell sheet suitable for cartilage repair. The present invention provides a cell sheet for cartilage repair, formed from a culture of cells derived from a cartilage tissue, and the cell sheet is negative for immunostaining using an antibody against type II collagen. The present invention also provides a method for producing a cell sheet for cartilage repair, formed from a culture of cells derived from a cartilage tissue, and the method includes culturing cells derived from a cartilage tissue on a surface of a membrane, where a temperature-responsive polymer is immobilized on the surface, to give the cell sheet. The culturing is stopped before the cell sheet becomes positive for immunostaining using an antibody against type II collagen.

摘要: Methods for ex vivo expansion of very small embryonic like stem cells in the absence of feeder cells are provided. In some embodiments the methods include providing a plurality of VSELs; and growing the VSELs in a culture medium that includes a histone deacetylase inhibitor, luteinizing hormone, follicle-stimulating hormone, and optionally transforming growth factor beta in an amount that is sufficient to overcome quiescence of the VSELs. Also provided are feeder cell-free cell cultures, ex vivo expanded VSELs, pharmaceutical compositions that include the disclosed ex vivo expanded VSELs, methods for overcoming quiescence in VSELs, methods for re-establishing imprinting in VSELs, method for treating injuries to tissues in subjects, methods for repopulating cell types in subjects, methods for bone marrow transplantation, methods for treating radiation exposure in subjects, and methods that relate to regenerative medicine.

摘要: The present disclosure provides methods of producing extracellular vesicles and methods of increasing extracellular vesicle production from producer cells, which exhibit a reduced gene and/or protein function in a cholesterol biosynthetic pathway. Also provided are cell compositions having a reduced gene and/or protein function in a cholesterol biosynthetic pathway. Reducing gene and/or protein function in a cholesterol biosynthetic pathway increases the yield and production of extracellular vesicles from the producer cells.

摘要: The present disclosure discloses a method for preparing a lysate, from a combination of umbilical cord blood (UCB) and maternal blood (MB) derived platelets by obtaining platelet-rich plasma via sedimentation processes and then mixing platelet-rich plasmas so obtained. The present disclosure further discloses a combination of umbilical cord blood and maternal blood platelet lysate (UCB+MB PL), and the PL so obtained shows greater efficacy as cell and tissue culture media supplement over and above its individual components, umbilical cord blood platelet lysate (UCB PL) and maternal blood platelet lysate (MB PL) and especially over foetal bovine serum (FBS). Moreover, the method of the present disclosure provides an economic, environment friendly, ethical and efficacious method to reach a cell and tissue culture supplement with far-reaching therapeutic applications.
