    Abstract: The present disclosure relates to a nucleic acid molecule including plural translation control elements downstream inserted downstream of a coding region, and a nucleic acid expression platform or a nucleic acid expression system such as a recombinant expression vector including the nucleic acid molecule inserted therein. A target gene inserted into the coding region can be expressed efficiently using the nucleic acid molecule. It is possible to improve expression efficiency of a report gene and, an antigen and/or therapeutic protein or peptide utilizing the expression platform.

    Abstract: This disclosure belongs to the field of biomedical technology, and particularly refers to a bifunctional molecule and the application thereof. The structure of the bifunctional molecule includes a first functional domain and a second functional domain. These domains are capable of simultaneously binding to T cells, thereby producing the first and second signals required for T cell activation. The bifunctional molecule is a recombinant protein-peptide, which can be produced by a eukaryotic cell expression system. The product has a single structure, simple purification process, high protein yield, and stable preparation process and product. The bifunctional molecule is superior to the current techniques in expanding T cells in vitro with lower protein dosage and simpler use procedure. It can be directly supplemented as soluble form without optimizing the relative ratio of full-length antibodies.

    Abstract: Provided are compositions and methods that include a CasY transactivating noncoding RNA (trancRNA) (referred to herein as a “CasY trancRNA”), nucleic acids encoding the CasY trancRNA, and/or a modified host cell comprising the CasY trancRNA (and/or a nucleic acid encoding the same). Subject compositions and methods can also include one or more of: (a) a “CasY” protein (also referred to as a CasY polypeptide, a Cas12d protein, and a Cas12d polypeptide), a nucleic acid encoding the CasY protein, and/or a modified host cell comprising the CasY protein (and/or a nucleic acid encoding the same); and (b) a CasY guide RNA (also referred to herein as a “Cas12d guide RNA”) that binds to and provides sequence specificity to the CasY protein, a nucleic acid encoding the CasY guide RNA, and/or a modified host cell comprising the CasY guide RNA (and/or a nucleic acid encoding the same).
