摘要: The present disclosure provides a Mycobacterium tuberculosis (Mtb) adenosine triphosphate (ATP) synthase-expressing recombinant bacterium, and a construction method and an expression method thereof, and belongs to the technical field of exogenous expression. The present disclosure provides a construction method of an Mtb ATP synthase-expressing recombinant bacterium, including the following steps: using a Mycobacterium smegmatis (Msm) competent cell containing an auxiliary gene knockout plasmid as a basal cell; transferring an Mtb ATP synthase gene cluster with an affinity purification tag into the basal cell, knocking out a Msm ATP synthase genome, and collecting a strain without the auxiliary gene knockout plasmid after conducting repeated subculture; and transferring the Mtb ATP synthase gene cluster into the strain by prokaryotic expression to obtain the Mtb ATP synthase-expressing recombinant bacterium.

摘要: The present invention provides a coupled enzyme system, comprises: a first enzyme, comprising a polyphenol phosphorylation synthetase; a second enzyme, which is ATP regeneration enzyme; and a substrate, being phosphorylated by the first enzyme. The coupled enzyme system of the present invention integrates polyphenol phosphorylation synthetase with ATP regeneration enzyme so that the polyphenol phosphorylation synthetase is used to phosphorylate polyphenol and the ATP regeneration enzyme regenerate ATP from AMP. Therefore, the present invention not only improves the water-solubility and bioavailability of the phenolic phytochemicals but also significantly reduces ATP consumption, presenting the potential of enzymatic systems in the production of polyphenol monophosphates.

摘要: A replication-competent recombinant virus includes an expression cassette encoding a protein associated with arginine biosynthesis. The protein associated with arginine biosynthesis may include argininosuccinate synthase 1 (ASS1), argininosuccinate lyase (ASL), or ornithine transcarboxylase (OTC). The recombinant virus may be derived from a myxoma virus. A method of treating a cell includes contacting a cell with a recombinant virus including an expression cassette encoding a protein associated with arginine biosynthesis. The protein associated with arginine biosynthesis may be ASS1, ASL, OTC.

摘要: The present invention relates to a method for producing eukaryotic cell lysates for cell-free protein synthesis comprising at least one exogenous enzyme, wherein the method comprises at least the following steps: a) providing eukaryotic cells transfected with at least one donor template coding for at least one exogenous enzyme which is selected from the group comprising orthogonal aminoacyl-tRNA synthetases and viral RNA polymerases: b) cultivating the transfected cells of step a) for a predetermined period of time and subsequently harvesting the cells; and c) disrupting the harvested cells and preparing a cell lysate for cell-free protein synthesis therefrom. More specifically, the invention relates to a method for producing eukaryotic cell lysates which are capable of cell-free synthesis of a target protein comprising a non-canonical amino acid, wherein the method comprises at least the following steps: a) providing eukaryotic cells transfected with at least one donor template coding for an orthogonal aminoacyl-tRNA synthetase. i.e. an aminoacyl-tRNA synthetase which is specific for a tRNA which is not recognized by endogenous aminoacyl-tRNA synthetase in said eukaryotic cells and is specific for a corresponding non-canonical amino acid: b) cultivating the transfected cells of step a) for a predetermined period of time and subsequently harvesting the cells; and c) disrupting the harvested cells and preparing a cell lysate therefrom. Further aspects of the present invention relate in particular to eukaryotic cell lysates obtainable by the above methods as well as to a method for performing cell-free synthesis of a target protein comprising a non-canonical amino acid.
