    Abstract: Multi-stage sample-recovery systems, including automated 2-stage and 3-stage sample-recovery systems, are provided. Such systems enable the rapid screening and recovery of samples, including viable cell-based samples, from high-throughput screening systems, including systems utilizing large-scale arrays of microcapillaries. In specific screening systems, each microcapillary comprises a solution containing a variant protein, an immobilized target molecule, and a reporter element. Immobilized target molecules may include any molecule of interest, including proteins, nucleic acids, carbohydrates, and other biomolecules. The association of a variant protein with a molecular target is assessed by measuring a signal from the reporter element. The contents of microcapillaries identified in the assays as containing variant proteins of interest can be identified and recovered using the multi-stage systems disclosed herein.

    Abstract: Disclosed are compositions and method related to variants of SPINK2 that bind to targets other than an endogenous target of SPINK2. In one embodiment, a peptide is provided that comprises the amino acid sequence SEQ ID NO: 1. In further embodiments, an amino acid sequences encoded by nucleotide positions 4 to 42 and/or nucleotide positions 94 to 189 in the nucleotide sequence of SEQ ID NO: 14 flank the amino terminus and the carboxyl terminus, respectively, of the amino acid sequence. In another embodiment, a peptide is provided that comprises an amino acid sequence derived from the amino acid sequence of SEQ ID NO: 1 in which a conservative substitution, deletion, 10 addition and/or insertion of 1 to 5 (inclusive) amino acids has occurred at amino acids other than the 1st X to the 12th X counting from the amino terminus.

    Abstract: Provided are methods and compositions for creating an arrayed library of cells. In particular, the methods and compositions provided herein improve the selection and transfer of cells by reducing contamination, increasing throughput, and improving consistency of transfer. Such methods and compositions include automated colony selection employing a solid barrier mounted on an automated apparatus for automatic handling of one or more pick tools. The solid barrier is configured to prevent undesired transfer of cells from a pick tool into the destination receptacle. Such methods and compositions also include acoustic transfer of cells in a high-density media by an acoustic liquid handler. The methods and compositions further include pooling the cells using a bitcode sample pooling scheme for high-throughput sequencing and deconvoluting the obtained nucleic acid sequences to identify the cells from which the sequences originated.

    Abstract: A method for synthesizing a synthetic molecule includes selecting a target protein, selecting a common precursor, providing first and second side-chains, forming a first monomer, including the common precursor and first side-chain, forming a second monomer, including the common precursor and second side-chain, evaluating the first and second monomers using an aggregation assay with the target protein to generate first results, and selecting one of the first and second monomers as a selected monomer based on the first results. Further, the method includes providing third and fourth monomers compatible with the selected monomer, forming a first dimer, including the selected monomer and third monomer, forming a second dimer, including the selected monomer and fourth monomer, evaluating the first and second dimers with the aggregation assay to generate second results, and selecting one of the first and second dimers as a selected dimer based on the second results.
