    Abstract: The present invention provides a novel adjuvant for polynucleotide vaccines, and in particular the present invention provides immunogenic compositions comprising a polynucleotide encoding an antigen capable of eliciting an immune response and an adjuvant comprising an immunostimulatory quantity of a gemini surfactant, or a derivative thereof. The invention further comprises polynucleotide vaccines that comprise, or are administered in association with, a composition that comprises a gemini surfactant compound. The polynucleotide vaccines of the present invention are vaccines that encode an antigen against which it is desired to generate an immune response, and in particular the polynucleotide vaccine may be a DNA vaccine. Also provided by the present invention is the use of gemini surfactants in the manufacture of a polynucleotide vaccine composition for the purpose of enhancing the immune response against the specific antigen that is encoded by the polynucleotide vaccine. Vaccine compositions, kits comprising separate polynucleotide composition and adjuvant compositions for separate or simultaneous administration, methods of manufacture of the vaccines and kits, and methods of treatment of individuals with the immunogencic compositions and vaccines of the present invention, are provided.

    Abstract: The present invention relates to efficient devices for administration of DNA based pharmaceutical agents into mammalian skin. In a particularly preferred aspect of the present invention there is provided devices for administration of DNA vaccines into the skin of the mammal, and preferably into human skin. The present invention provides a microneedle DNA pharmaceutical agent delivery device having at least one skin-piercing element which comprises a support member coated with a solid reservoir medium containing, in solid solution or suspension within it, the DNA pharmaceutical agent, and a stabilising agent that inhibits the degradative effects of free radicals. Preferably the solid pharmaceutical reservoir medium coated onto such devices is a polyol, preferably being a polyol in an amorphous state (such as a glass).

    Abstract: The present invention relates to novel compositions comprising DNA attached to an immunostimulatory oligonucleotide (CpG) via a locked nucleic acid oligonucleotide of a defined sequence. In particular the present invention provides compositions comprising a plasmid containing a gene encoding a protein of interest, wherein said plasmid may be introduced to a tissue or cell and the gene expressed, complexed to the LNA -CpG. Also provided is a method of co-synthesis of CpG and LNA to form compositions for use as adjuvants with DNA plasmid vaccines.
