    摘要: A new method and system for desorption ionization is described and applied to the ionization of various compounds, including peptides and proteins present on metal, polymer, and mineral surfaces. Desorption electrospray ionization (DESI) is carried out by directing charged droplets and/or ions of a liquid onto the surface to be analyzed. The impact of the charged particles on the surface produces gaseous ions of material originally present on the surface. The resulting mass spectra are similar to normal ESI mass spectra in that they show mainly singly or multiply charged molecular ions of the analytes. The DESI phenomenon was observed both in the case of conductive and insulator surfaces and for compounds ranging from nonpolar small molecules such as lycopene, the alkaloid coniceine, and small drugs, through polar compounds such as peptides and proteins. Changes in the solution that is sprayed can be used to selectively ionize particular compounds, including those in biological matrices. In vivo analysis is demonstrated.

    摘要: The present invention provides a method for detecting a target analyte in a sample, the method comprising:
    introducing a charged mass label composition to a sample comprising a target analyte, wherein the charged mass label composition comprises an affinity reagent; and a mass label precursor bound to the affinity reagent, wherein the mass label precursor comprises a label binding unit and a mass label, the label binding unit reversibly binding the mass label to the affinity reagent, and wherein the mass label comprises a charge unit and a mass label unit having a pre-defined mass-to-charge-value in a mass spectrum;
    incubating the sample and the charged mass label composition so that the charged mass label composition binds the target analyte via an interaction between the affinity reagent of the charged mass label composition and the target analyte;
    releasing the mass label from the affinity reagent by breaking one or more bonds of the label binding unit;
    ionizing the mass label; and
    detecting the ionized mass label and/or fragments thereof in a mass spectrometer, thereby detecting the target analyte in the sample.

    摘要: An improvement to Desorption Electrospray Ionization (DESI), the process of creating ions directly from sample surfaces for mass spectrometric (MS) analysis by impinging a liquid spray onto the surface. The improvement is brought about by enclosing the spray and sample surface and MS-inlet capillary in a pressure tight enclosure. The invention includes methods of sampling a larger or smaller area of surface by impacting and collecting droplets from such an area. The invention allows DESI to be performed without need for careful control of the geometry of the sprayer and MS-inlet capillary positions and angles relative to the sample surface.
