摘要:
The present invention is directed to a monoclonal antibody or a fragment thereof capable of binding to at least one epitope of the α-subunit of the human high affinity Fc IgE receptor, hybridoma cells secreting such a monoclonal antibody, a process for the preparation of such a hybridoma cell and for the preparation of such a monoclonal antibody or fragment thereof, pharmaceutical compositions containing such an antibody or fragment thereof and the use of such an antibody or fragment thereof for therapy, diagnosis and purification purposes.

A diagnostic site-specific imaging reagent in the form of a receptor and an indicating means selectively binds to a specific cell membrance-associated antigen of a blood platelet that is in a stimulated active state but does not substantially bind to a platelet that is in a non-stimulated resting state. In particular, prelabelled monoclonal antibodies and polyclonal antibodies are prepared that bind and image thrombospondin when it is cell membrance-associated on a thrombin-stimulated platelet, thereby providing means for detecting and discriminating between a stimulated active blood platelet and a non-stimulated resting blood platelet.

The present invention first provides monoclonal antibodies recognizing membrane phospholipase A₂, namely, monoclonal antibodies PL-49, PL-71, PL-76, and PL-78, hybridomas producing them, methods for producing them, and immunoassays of membrane phospholipase A₂ using them. The immunoassay of PLA₂M is useful for the diagnosis of articular rheumatism, cancers, and a wide variety of inflammatory states.

A method of producing rat monoclonal antibodies to human T-cell lymphocyte antigen epitopes other than those epitopes defined by murine monoclonal antibodies directed to the same antigen comprising: (1) immunizing a rat or rat immunocompetent cells in vitro with an immunogenic amount of an antigen comprising T-cell lymphocyte markers; (2) fusing immunized cells from said rat or immunized rat immunocompetent cells with immunocytoma cells; (3) selecting hybridoma cells that produce antibody that binds to the human T-cell lymphocyte antigen epitopes other than those epitopes defined by murine monoclonal antibodies directed to the same antigen; (4) culturing the selected hybridoma cells; and (5) recovering said antibody. A method of producing rat monoclonal antibodies that bind to human CD4 antigen, the method comprising: (1) immunizing a rat or rat immunocompetent cells in vitro with an immunogenic amount of an antigen comprising CD4 molecule; (2) fusing immunized cells from said rat or immunized rat immunocompetent cells with immunocytoma cells; (3) selecting hybridoma cells that produce antibody that binds to human CD4 antigen; (4) culturing the selected hybridoma cells; and (5) recovering said antibody. A hybridoma that produces a rat monoclonal antibody that binds to human CD4 antigen. A rat monoclonal antibody that binds to human CD4 antigen. In a preferred embodiment the rat monoclonal antibody binds to a human CD4 antigen epitope other than those epitopes defined by murine monoclonal antibodies OKT4 and Leu3a. A rat monoclonal antibody that binds to human T-cell lymphocyte antigen epitopes other than those epitopes defined by murine monoclonal antibodies directed to the same antigen.

A novel established cell lint (ECACC 86 091801) is obtained by collecting lymphoid cells from an animal with autoimmune disease, culturing them in a culture medium containing the supernatant of a culture in which lymphoid cells of animal origin have been grown in the presence of a mitogen, and continuing the culture for a period of 8 months until the cells come to grow in the absence of the aforesaid supernatant. This established cell line has the following properties: (1) it produces a factor participating in the differentiation of antibody-producing cells. (2) it does not produce B cell growth factor. (3) it consists of immortalized null cells. This established cell line can be used to enhance the production of anti-nucleic acid antibodies by antibody-producing cells.

Provided in the present invention are an anti-OX40 monoclonal antibody or an antigen-binding fragment thereof, and a corresponding isolated polynucleotide, expression vector, recombinant cell, pharmaceutical composition, and application thereof. The present antibody can be used for the treatment of cancer or autoimmune disease.
