The present invention provides novel processes for the release of vitamin B₁₂ and/or folate in mammalian serum samples from endogenous serum binders comprising microwaving the serum samples prior to assaying the serum samples for vitamin B₁₂ and/or folate. Prior to the assay procedure, the serum samples are typically microwaved for about 20-90 seconds, most preferably for about 60 seconds at a high setting or such that the temperature of the serum samples is brought to a temperature of about 65 to 100°C, most preferably 80-85°C, for at least 15 seconds.

The present invention relates to compositions and methods for the detection of sperm antibodies bound to the surface of spermatozoa or present in human serum, seminal plasma or cervical mucous through the use of immunoassays employing latex beads coupled to anti-human immunoglobulins. Preferably the beads are monodispersed polystyrene beads coupled to the anti-human immunoglobulins through a bridge of a polyamine, such as polylysine, having amino acid residues of about 200 to about 1,000. The beads are also preferably about 2 to about 4 microns in size.

An immunoassay utilizing a biotin bridge is performed with a solid phase with anti-biotin immobilized on its surface. In accordance with the assay, the solid phase is incubated with a biotinylated binding species, preferably biotinylated antibody, specific for the ligand to be detected. This is then reacted with the test sample, followed by a labeled binding member, either in a sandwich-type or competitive-type binding assay. The label is then detected in accordance with conventional detection techniques. Use of the solid phase with only a biotin-specific antibody attached thereto expands the utility and versatility of a single solid phase for use in the detection of an unlimited variety of ligands.
