摘要： Provided herein is a bispecific antibody and the use thereof. The bispecific antibody comprises an antibody or antibody fragment; the antibody or the antibody fragment is a bispecific antibody or an antibody fragment comprising a first heavy chain, a first light chain, a second heavy chain, and a second light chain; the first Fab region comprises the first heavy chain and the first light chain; the second Fab region comprises the second heavy chain and the second light chain; and a variety of mutations are introduced in the first Fab region and the second Fab region. By means of performing amino acid size and charge mutations on amino acid residues at a heavy chain CH1 and light chain CL interface of the antibody or the antibody fragment, the correct pairing of heavy/light chains targeting different antigens is enhanced, the binding activity to specific antigens is maintained, mismatches are reduced, the binding specificity is improved, and the yield of a target product is improved.
