Disclosed are modified lipase enzymes, as well as polynucleotides and recombinant expression vectors encoding the lipase variant polypeptides, as well as methods for producing such lipase variants in selected bacterial and fungal host cells. The invention further relates to lipase variants having a preference for medium- or long- chain fatty acid esters or carboxylic acid moieties with carbon-chain lengths greater than or equal to C 6 . The invention further relates to methods for selectively reducing, or eliminating, one or more trans-fatty acids from substrates, including liquid edible oils useful in food manufacture and frying systems.

The present invention relates to a newly-identified lipase enzyme isolated from a member of the Ustilaginaceae family of the Basidomycetes , and to polypeptide variants thereof, that exhibit increased trans -selectivity, and/or increased preference for catalysis of long-chain fatty acid esters or carboxylic acid moieties having chain lengths greater than twelve carbons (C 12 ). The invention also provides polynucleotides and nucleic acid sequence that encode one or more lipases and/or variants thereof, and to recombinant expression vectors, and host cells that comprise them. The invention also provides methods for producing the lipase, and variants thereof both in vivo , and recombinantly in vitro , including large-scale commercial processes suitable for use in the food and cosmetics industries. Methods are also provided for altering, reducing, and/or eliminating one or more trans-fatty acids from cooking oils, including frying oils, and related products.
