The present invention provides a novel procedure of preparing samples for analysis by way of mass spectrometry, preferably LC-MS/MS. Accordingly, functionalized magnetic particles with a hydrophobic surface are used for extracting low molecular weight compounds from complex liquid biological samples such as plasma, serum, whole blood or hemolyzed blood. The method of the invention includes (a) contacting the sample with an amount of functionalized magnetic particles with a hydrophobic surface, (b) incubating the sample and the particles, thereby adsorbing the compound to the hydrophobic surface, (c) separating the particles by applying a magnetic field and removing the liquid, (d) optionally washing the particles, (e) eluting the compound from the particles.

In a section (97) of the extractor (90) the microparticles are immobilized by means of a magnetic field of a controllable means (96) and separated from the remaining sample material. By switching over of a switching unit (110) a second mobile phase (75) is carried to the extractor (90) and the second mobile phase (75) detaches the adsorbed analyte from the surface of the microparticles. Gegenstand der Erfindung ist eine Vorrichtung und ein Verfahren zur Manipulation von einem flüssigen Probenmaterial und darin suspendierten magnetischen Mikropartikeln, an deren funktionalisierter Oberfläche Analyten gebunden sind. Das Probenmaterial wird durch eine Injektionsvorrichtung (50) in ein Flüssigkeitssystem eingebracht und von einer ersten mobilen Phase (65) einem Extraktor (90) zugeführt. Die Mikropartikel werden im Extraktor (90) in einem ersten Abschnitt (97a) durch ein Magnetfeld einer steuerbaren Vorrichtung (96) immobilisiert und vom übrigen Probenmaterial getrennt. Durch Umschalten einer Schalteinheit (110) wird in den Extraktor (90) eine zweite mobile Phase (75) geleitet, die die adsorbierten Analyten von der Oberfläche der Mikropartikel löst. Die Mikropartikel werden in einem zweiten Abschnitt (97b) der steuerbaren Vorrichtung (96) immobilisiert und die in der zweiten mobilen Phase (75) gelösten Analyten können mittels chromatographischer Trennung (130) und anschliessender Detektion (140) nachgewiesen werden.
