wherein the immunostimulatory oligonucleotide includes a modified backbone, at least one nucleotide separates consecutive CpGs, X 1 X 2 is GpT or ApT, N 1 is any nucleotide that includes a 5' TC or TG, N 1 + N 2 is from 0-26 bases, and the immunostimulatory oligonucleotide is from 8-30 bases in length and begins with TC or TG at the 5' end, with the proviso that neither N 1 nor N 2 contains a CCGG quadmer or more than one CCG or CGG trimer.

These nucleic acids containing at least one unmethylated cytosine-guanine (CpG) dinucleotide can be used to induce an immune response in a subject. Methods and compositions are provided for extending the clinical utility of IFN-α in the treatment of a variety of viral and proliferative disorders. Among other aspects, the invention provides methods which increase the efficacy of IFN-α treatment and reduce IFN-α treatment-related side effects. In addition, methods are provides for supporting the survival and for activating natural interferon producing cells (IPCs) in vitro without exogenous IL-3 or GM-CSF. The invention is based on the discovery that certain CpG and non-CpG ISNAs promote survival and stimulation of IPCs.
