摘要： PURPOSE:To prepare D-aminoacylase by culturing facultative methanol-assimilating bacteria. CONSTITUTION:A strain selected from pseudomonas aminovorans NCIB 9039, Pseudomonas sp. 1158 (FERM-P No.4632) and Pseudomonas sp.617 (FERM-P No.4631) is cultured in a conventional medium at 10-45 deg.C and pH 4.0-9.0 to obtain the objective D-aminoacylase. The characteristics of the aminoacylase are as follows: (1) substrate specificity, hydrolyzes the acyl group of N-acyl-D-amino acid, and inactive to N-acyl-glucosamine or N-acyl-ethanolamine; (2) stability, keeps its activity at 50 deg.C, most stable at pH 6-7; (3) molecular weight, 100,000; (4) isoelectric point, pH=4.95; (5) elemental analysis, C 54.33%, H 7.19% and N 16.37%; (6) inhibitor, remarkably inhibited by p-chloro-mercury benzoic acid., etc.
