PROBLEM TO BE SOLVED: To provide improved fed batch methods and compositions for promoting protein expression in cell culture, particularly mammalian cell culture.SOLUTION: Improved salt-free basal growth media for use in mammalian cell culture includes a serum-free cell culture medium comprising Part A, Part B, and Part C, wherein Part A consists essentially of a modified basal medium which excludes the following components: sodium bicarbonate, a buffer, sodium phosphate, dibasic sodium phosphate, an osmolality regulator, a surfactant, and monosaccharide glucose; Part B consists essentially of an inorganic iron source; and Part C comprises a recombinant growth factor; a buffer; an osmolality regulator; an energy source; and at least two different non-animal hydrolysates. The fed batch method for culturing cells includes adding a hydrolysate concentrated solution and a basal concentrated solution to a cell culture during a time period.

PROBLEM TO BE SOLVED: To provide a method of efficiently producing EPO having similar properties to naturally occurring EPO; and to provide a method of efficiently isolating EPO-producing cells.SOLUTION: The present invention provides a method for producing erythropoietin, comprising culturing neural crest-derived cells in a medium to allow production of erythropoietin and recovering erythropoietin from the medium. The present invention also provides a method for isolating erythropoietin-producing cells from a renal tissue, comprising isolating erythropoietin-producing cells from a cell population contained in the renal tissue using a neural crest-derived cell-specific surface antigen(s).
