摘要: Provided herein is a method for producing a population of symmetrically barcoded transposomes, i.e., transposomes that are loaded with a pair of identical double-stranded adapters. In some embodiments, this method may comprise amplifying a template that contains a randomized sequence and a transposon end sequence on a solid support by bridge polymerase chain reaction (PCR) to produce uniquely barcoded clusters of single-stranded amplification products that are tethered to the support, processing the single-stranded amplification products so that the transposon end sequence is double-stranded and at the end of the products, and adding transposase to the support under conditions by which the transposase binds to the double-stranded transposon end sequences, to produce population of symmetrically barcoded transposomes.
