摘要： Provided is a method for preparation of a composition comprising activated human CD8+ lymphocytes with phenotype of stem cell-like memory cells and natural killer (NK) lymphocytes. The method entails use of short-term activation of lymphocytes by CD3/CD28 activating agents followed by treatment with DNA-demethylating agent. The invention also provides a version of the method where addition of a CD3/CD28 activating agent is made a few days after initiation of CD4+ mediated activation of the CD8+ cells; this step is also disclosed as an improvement of related methods where autologous dendritic cells have been used to activate the lymphocytes. Also provided is a method for treatment of cancer using the cells obtained from the process.

摘要： A method of cultivation of cells comprises use of a vessel (1;1';11;17) having at least one interior surface (3;14;21) provided with a plurality of parallel grooves (4); said grooves being arrange for cultivation of cells. A simple tilting regime of the vessel provides the possibility of either exchanging the media or gently harvesting the cells, depending on the tilt direction of the vessel in relation to the longitudinal axis of the grooves.

摘要： The present invention relates to compositions and methods for producing dendritic cells and particularly to compositions and methods for producing immature dendritic cells that are immunocompetent. We describe a method of producing dendritic cells by cultivation of monocytes, characterised by at least one of: pre-treatment of a tissue culture surface with at least one of: a substantially plasma-free and serum-free pre-treatment medium, a pre-treatment medium comprising heparin, and a pre-treatment medium comprising a protein solution; adsorption of monocytes using at least one of: a substantially plasma-free and serum-free adsorption medium; cultivation of monocytes using a substantially plasma-free and serum-free cultivation medium. We also describe compositions including the dentritic cells and uses of the dentritic cells.

The method entails use of allogeneic mature dendritic cells as feeder cells added at an early stage in the induction of proliferation and activation of CD4+ and CD8+ T cells.
