The compositions include pharmaceutical preparations that contain isolated or recombinant or modified peptidase D (PEPD) proteins. The methods include prophylaxis and/or therapy of ErbB2-positive cancer by administering a PEPD to an individual who has or is at risk for developing ErbB2- positive cancer.

The invention provides an isolated, novel steroid 5α-reductase enzyme termed SRD5AIIL The protein has an estimated molecular weight of 37 kDa and is capable of converting testosterone to dihydrotestosterone at a pH of about 7.0. Also provided is a method for identifying inhibitors of SRD5 AIII by contacting SRD5 AIII with a test compound and measuring the activity of the enzyme. A reduced activity relative to a control indicates that the test compound is an inhibitor of SRD5AIII. A method is also provided for detecting androgen stimulated prostate cancer or recurrent prostate cancer in an individual. The method comprises obtaining a prostate biopsy from an individual and determining the level of expression of SRD5ADI gene or protein relative to a normal control. An increased expression of SRD5AIII relative to the control is indicative of androgen stimulated prostate cancer or recurrent prostate cancer.

A method for detecting circulating levels of beta amyloid peptides in an individual by administration of one or more inhibitors of an extracellular proteolytic pathway in blood and detection of the beta amyloid peptides in the blood or a fraction thereof. An example of an inhibitor is an anticoagulant. Levels of desired beta amyloid peptides such as Αβ1-40 and Αβ1-42 can be determined in the blood samples collected from the individuals.

Provided are methods for prophylaxis or therapy of cancer. The methods are directed to cancers that are characterized by expression of a mutant p53. The mutant p53 may be a loss of function p53 mutant, dominant negative p53 mutant, or a gain of function p53 mutant. The method comprises delivering to cancer cells an agent that can inhibit expression of prolidase (PEPD) or disrupts the association of mutant p53 with PEPD.

Provided is a composition that contains a mixture of brocolli seed and mustard seed. Before mixning the brocolli seed with the mustard seed, the broccoli seed is subjected to baking and a pressurized heat treatment. The broccolli seed is baked at a temperature of at least 200 degrees Fahrenheit for at least 60 minutes, and is subjected to a pressurized heat treatment of at least 200 degrees Fahrenheit at a pressure of at least 10 pounds/square inch for at least 5 minutes. Also provided is a method for therapy and/or prophylaxis of bladder cancer in an individual The method entails administering orally to the individual a composition that contains an isothiocyanate (ITC) such that the administration inhibits the growth and/or recurrence of bladder cancer.
