A bubble maker simplifies the creation and distribution of bubbles. The bubble maker includes a mixing basin, a water-inlet tube, an air-inlet tube, a bubble outlet, and a water overflow outlet. The bubble maker does not require an external electric power source to drive the agitation for a mixture of the water and soap as Venturi effect is implemented via water pressure from an external water supply through the water-inlet tube into the air-inlet tube to aerate the water and soap mixture and produce bubbles. The bubble maintains a maximum water level within the mixing basin with a water overflow outlet that allows for sufficient headspace for bubbles to accumulate within the mixing basin. As the bubbles accumulate within the mixing basin, the bubbles flow out of the bubble maker through the bubble outlet.

It has been discovered that NAD+-dependent histone deacetylase SIRT6 is critical for suppression of PDAC by controlling the expression of Lin28b, which is a negative regulator of let-7 microRNA. Specifically, SIRT6 loss results in histone hyperacetylation at the Lin28b promoter, Myc recruitment, and pronounced induction of Lin28b and downstream let-7 target genes, HMGA2, IGF2BP1, and IGF2BP3. This invention relates generally to agents and methods of reducing expression or activity of Lin28b to treat (aggressive) PDAC in a subject.

This invention is based on the discovery that many eye conditions associated with aging are mediated at least in part by cells bearing a senescent phenotype. Senescent cells accumulate with age, and express factors that contribute to the pathophysiology of age related conditions. The data show that in age-matched patients, the severity of age-related conditions correlates with the abundance of senescent cells, and that clearing senescent cells can help abrogate the condition. Small molecule drugs that remove senescent cells from affected tissue in the eye are provided that have special efficacy in treating ophthalmic conditions. They not only inhibit progression of the disease, they can also reverse some of the pathophysiology—such as neovascularization and vaso-obliteration—that lead to vision loss. These senolytic agents have an appropriate dose and specificity profile to be effective in the clinical management of previously intractable ophthalmic conditions.
