It relates to a method for amplifying at least one nucleotide molecule contained in a sample (E) comprising the following successive steps consisting of (a) subjecting said sample (E) to a 1st amplification step on an area (Z1) of a support; (b) bringing by electrowetting at least one portion of the sample (Ea) obtained after step (a) from the area (Z1) onto at least one area (Z2) of said support, distinct from the area (Z1); (c) subjecting the sample (Ea) to a 2nd amplification step on said area (Z2). It also relates to a device which may be applied within the scope of said method.
