The present invention provides methods of multi-dimensional profiling of biologically active agents and determining their effects on biological systems. The methods of the present invention include real-time impedance monitoring of cellular responses to biologically active agents and categorization of cellular kinetic profiles into mechanism specific cellular response profile groups. The grouping of similar cellular response profiles allows the correlation between agent and mechanism, thus allowing for the identification of potential therapeutic applications of agents or further study of cellular responses or mechanisms.

Abstract:
A device for monitoring the migration or invasion of a biological particle such as a cell is disclosed. The device includes an upper chamber adapted to receive and retain a cell sample, a lower chamber having at least two electrodes, and a biocompatible porous membrane having a porosity sufficient to allow cells to migrate therethrough. The membrane is disposed in the device so as to separate the upper and lower chambers from one another. Migration of cells through the porous membrane permits contact between the migrating cells and one or more electrodes of the lower chamber. The contact provides a detectable change in impedance between or among the electrodes.

Methods of assessing cytolysis of cancer cells, including providing a cell-substrate impedance monitoring device operably connected to an impedance analyzer, wherein the device comprises a well for receiving cells and an electrode array at a base of the well; adding target cells characterized as cancer cells to the well; adding effector cells to the well to form a test well, wherein the effector cells are immune cells obtained or derived from a same patient as the target cells; monitoring cell-substrate impedance of the test well before and after adding the effector cells and optionally deriving an impedance-based parameter from the impedance; and determining effectiveness of effector cell killing of the target cells by comparing the impedance or impedance based parameter over time.
Abstract:
Novel compounds having a fused bicyclic heteroaromatic ring system substituted with a heteroaryl five-membered ring are disclosed. The compounds inhibit growth of a variety of types of cancer cells, and are thus useful for treating cancer. Efficacy of these compounds is demonstrated with a system for monitoring cell growth/migration, which shows they are potent inhibitors of growth and/or migration of cancer cells. In addition, compounds of the invention were shown to stop growth of tumors in vivo, and to reduce the size of tumors in vivo. Compositions comprising these compounds, and methods to use these compounds and compositions for treatment of cancers, are disclosed.
