A method of minimizing dead-periods in magnetic resonance imaging pulse sequences employs a specimen disposed within magnetic field (6), a source of RF signals (2), a receiver (8) for receiving signals from the specimen responsive to RF pulses and emitting respective output signals. A computer (12) is provided for receiving the output signals from the receiver and establishing image information which may be displayed (16). For the dead-period, the minimum and maximum phase encoding step, the scan plane gradient pulse for the slice, phase encoding and readout directions are determined and the moments contained within the dead-period waveform is determined. The values are transformed into gradient amplifier coordinates and the minimum dead-period based on the dead-period waveform is determined. The dead-period is employed in establishing a hardware optimized waveform which may be trapezoidal. The trapezoidal waveform is preferably established by for each phase encoding step determining the starting and ending gradient levels and the moments contained within the dead-period waveform using the calculated minimum dead-period.

A novel protein tyrosine kinase, JAK3, and a polynucleotide sequence encoding JAK3 polypeptide are disclosed herein. JAK3 is a new member of the JAK family of protein tyrosine kinases which are important in regulation of cellular proliferation and differentiation. Also disclosed are therapeutic methods utilizing JAK3 polypeptide and polynucleotide sequences.

Methods for separating target particles from a test medium in which the particles are suspended. The method contemplates utilization of blocking or masking particles which block or mask unwanted biological debris which tends to interfere with formation of complexes by which the target particles can be isolated. After centrifugation, the masking particles create a barrier between unwanted debris and test medium containing the target particles which permits the particles to be manipulated or modified without removing them from the debris. In a preferred aspect, magnetic separation techniques are used. In this aspect, non-magnetic 0.04 micron carboxylated polystyrene blocking particles are employed.

Serial analysis of gene expression (SAGE) was used to identify transcripts encoding secreted or cell-surface proteins that were expressed in benign and malignant tumors of the colorectum. A total of 290,394 tags were analyzed from normal, adenomatous and cancerous colonic epithelium. Of the 21,343 different transcripts observed, 957 were found to be differentially expressed between normal and adenoma or between normal and cancer. Forty-nine transcripts were elevated ≥ 20-fold in adenomas, 40 transcripts were elevated ≥ 20-fold in cancers, and nine transcripts were elevated ≥ 20-fold in both. Product of six these nine transcripts (TGFBI, LYS, RDP, MIC-1, REGA, and DEHL) were predicted to be secreted or to reside on the cell surface and these were analyzed in more detail. The abnormal expression levels predicted by SAGE were confirmed by quantitative PCR analyses of each of these six genes. Moreover, the cell types responsible for the elevated expression were identified by in situ hybridization and by PCR analyses of epithelial cells immunoaffinity purified from primary tumors.

A novel T-type calcium channel (CACNA1G) is provided, as are polynucleotides encoding the same. CACNA1G has been implicated in cellular proliferative disorders. More specifically, it has been observed that the methylation state of specific regions within CpG islands associated with the CACNA1G gene correlates with a number of cancerous phenotypes involving a variety of tissue and cell types. Also provided are methods for detecting cellular proliferative disorders by determining the methylation state of genes or regulatory regions associated therewith, including CACNA1G, as well as kits containing reagents for performing invention methods.
