The invention provides Sf2-deficiency or prophylactic administration of a therapeutic agent, such as a Sf2 antagonist or a Sf2 antibody, markedly reduces thrombus formation that would otherwise be caused due to surgery, immobility, genetic abnormality, cancer, trauma, or other risk factors.

Isolated nucleic acid molecules encoding monoclonal antibodies that bind TFPI are also contemplated. Pharmaceutical compositions comprising the anti-TFPI monoclonal antibodies and methods of treating deficiencies or defects in coagulation by administration of the antibodies are also provided. Methods of producing the antibodies are also provided.

This disclosure provides pro-drug antibodies which specifically bind to tissue factor pathway inhibitor (TFPI) only after exposure to proteases from the coagulation cascade. The pro-drug antibodies described are useful for treating bleeding disorders such as hemophilia. When used in such treatments, these pro-drug antibodies show increased half-life relative to other anti-TFPI antibodies, while mitigating the potential for side effects such as thrombosis.

Methods of using antibodies to modulate PAI-1 activity or detect PAI-1, either in vitro or in vivo, are also provided.
