The syringe comprises a body (20, 21, 22) having a lumen with a diaphragm (26) located adjacent to a terminus thereof. Particles are delivered from an external surface of the diaphragm (26) by means of the motive force provided by the impact of a shockwave imparted to the internal surface of the diaphragm. 摘要: The invention provides a silencer and silencing method which ensures adequate silencing with no deleterious increase in back pressure (which creates a lift-off force) or decrease in device performance. Pressurised gas is supplied to a driver chamber by a bleed-hole having an effective bleed-hole area and, during use of the device for particle delivery, gas is vented to the atmosphere via a silencer having an effective venting area. The bleed-hole area and venting area are chosen to ensure that the mass flow rate of gas through the effective venting area is substantially equal to or greater than the mass flow rate of gas through the effective bleed-hole area. This ensures that there is no build up of gas in the silencer device which tends to increase back pressure and hence lift-off force. Preferred embodiments of the device comprise a silencer having a large volume and a small particle exit opening. Further, there is disclosed an embodiment using one or more transfer ducts to assist particle mixing.

摘要: There is disclosed a method of distributing particles in a flow of gas and a needleless syringe for use in the needleless injection of particles into the skin or mucosa of a vertebrate subject. The syringe has the advantage that no bursting membranes are required, reducing noise, and that the particles are substantially uniformly distributed in the gas stream. This is achieved by providing a convergence which reduces pressure of the gas flowing in the gas flow path due to the Venturi effect such that particles initially located outside of the gas flow path are drawn into the gas flow path under the action of the reduced pressure and become entrained in the gas. An exit nozzle is provided to accelerate the particles so entrained. In another aspect of the invention, there is provided a method of creating a gas flow in a needleless syringe which comprises flowing gas through a first convergence into a chamber to form a transsonic gas jet in the chamber and passing the gas jet from the chamber into a second convergence and along the nozzle. This provides an advantageous flow regime suitable for the needleless injection of powders.

摘要: The invention provides mechanisms to improve particle atomisation in a needleless syringe. If the particles are more evenly distributed across the cross-section of the device at the time at which they are entrained, a better footprint of particle penetration is obtainable. The particles are initially located between first and second closure means and the pressure at which said closure means open can be made to be different. Also, a transfer duct which fluidly connects a location upstream of the second closure means to a location between the first and second closure means can be provided. Furthermore, the closure means can be constituted by rupturable membranes which are scored or indented so as to provide controlled rupturing.

摘要: Nucleic acid constructs comprising viral genomic nucleic acid which comprises at least two endogenous gene expression regulatory units which each comprise an endogenous promoter where the endogenous promoters of the units are active at the same phase in the viral life cycle of the virus the viral genomic nucleic acid is derived from are provided. The endogenous gene expression regulatory units are used to express particular chosen heterologous coding sequences and in particular to express heterologous antigens. The constructs can be used in a vaccine to generate an immune response against the heterologous antigens and in particular may be used in a DNA vaccine. Methods for generating the constructs and means for their administration are also provided.

摘要: The invention provides a device which seeks to ensure that substantially all the particles delivered avoid interaction with the so-called 'starting process'. There is provided a needleless injection device comprising a driver chamber (51) arranged, in use, to contain a charge of pressurised gas, a duct section (52) connected to the driver chamber (51) to receive gas therefrom and a closure means (53) for preventing the flow of gas from the driver chamber (51) to the duct section (52) until the closure means (53) is opened. Further, a dose of particles (58) is positioned within the device in the region of the closure means (53). The device is so constructed and arranged that upon opening of the closure means (53), a primary shock wave is produced to travel along the duct (52) in a downstream direction so that a substantially quasi-steady gas flow is established in the duct (52) upstream of the primary shock wave, with the dose of particles (58) being substantially wholly entrained in the substantially quasi-steady flow to be accelerated thereby and expelled from the device.
