Gas or air filled microbubble suspensions in aqueous phases usable as imaging contrast agents in ultrasonic echography. They contain laminarized surfactants and, optionally, hydrophilic stabilizers. The laminarized surfactants can be in the form of liposomes. The suspensions are obtained by exposing the laminarized surfactants to air or a gas before or after admixing with an aqueous phase.

On décrit des suspensions injectables de microbulles de gaz dans un vecteur liquide aqueux utilisables comme agents de contraste dans l'échographie à ultrasons. Les composites contiennent des agents amphiphatiques dont au moins un peut être un phospholipide à structure lamellaire, comme stabilisants des microbulles pour éviter leur coalescence sous l'effet du temps et de la pression. La concentration en phospholipides dasn le liquide vecteur est inférieure à 0,01 % en poids, mais elle est au moins égale ou supérieure à celle où les molécules de phospholipides sont uniquement présentes à l'interface microbulles-liquide. On décrit également unprocédé de préparation de suspensions stables de microbulles constituées d'air ou d'un gaz.

A solution is proposed for analyzing a body part (155) of a patient (160), which is perfused with a contrast agent capable of circulating within the patient and of being substantially immobilized on a biological target. A corresponding system (105) includes means (405-440) for providing an echo signal indicative of a response over time to an interrogation signal of the body part, means (442,450) for associating the echo signal with a model function of time modeling an evolution over time of the contrast agent in the body part, the model function including a combination of a circulation function modeling the circulation of the contrast agent and a decay function modeling a decay of the echo signal from the immobilized contrast agent, and means (455-475) for estimating at least one kinetics indicator of the contrast agent from the model function.

Method for preparing a lyophilized matrix and, upon reconstitution of the same, a respective injectable contrast agent comprising a liquid aqueous suspension of gas-filled microbubbles stabilized predominantly by a phospholipid. The method comprises preparing an emulsion from an aqueous medium, a phospholipid and a water immiscible organic solvent. The emulsion is then freeze-dried and subsequently reconstituted in an aqueous suspension of gas-filled microbubbles. The method allows to obtain suspensions comprising microbubbles having a relatively small diameter and a narrow size distribution.

Disclosed are injectable suspensions of gas filled microbubbles in an aqueous carrier liquid usable as contrast agents in ultrasonic echography. The suspensions comprise amphipathic compounds of which at least one may be a laminarized phospholipid as a stabiliser of the microbubbles against collapse with time and pressure. The concentration of phospholipids in the carrier liquid is below 0.01 % wt but is at least equal to or above that at which phospholipid molecules are present solely at the gas microbubble-liquid interface. Also disclosed is a method of preparation of the stable suspensions of air or gas filled microbubbles.

Liposome vesicles are prepared containing water or very dilute solutions encapsulated therein. These 'empty' liposomes are suspended in a carrier liquid containing, dispersed therein, substances of interest to be loaded into the vesicles and incubated for a period of time at temperatures above the lipids transition temperature, whereby loading by transmembrane permeation occurs in high yields.
