摘要： By screening a swine genomic DNA library, a BAC clone containing a swine RAG-1 gene is obtained. From this BAC clone, RAG-1 gene-containing regions are subcloned to give 3 clones. These clones are sequenced to determine the base sequences and mapped by digesting with restriction enzymes, thereby form gene maps. Based on the thus obtained data, a targeting vector for knockouting the RAG-1 gene is constructed. By regulating the function of the endogenous swine RAG-1 gene with the use of this vector, a swine having no acquired immunity can be constructed.
