This invention provides a method of determining an immune complex specific to a tumor associated antigen characterized in that it comprises the steps of:
(1) reacting a complement-binding, tumor associated antigen-antibody complex present in a body fluid as an immune complex either with an insolubilized anti-complement or with an insolubilized substance specifically bindable to the tumor associated antigen selected from the group consisting of basic fetoprotein (BFP), a-fetoprotein (AFP), tumor associated glycolinkage (TAG), and carcinoembryonic antigen (CEA) or the antibody thereagainst constituting the complex to form an immune complex-insolubilized anti-complement bound product or an immune complex-insolubilized substance bound product. (2) reacting the immune complex-insolubilized anti-complement bound product with a predetermined amount of labelled substance specifically bindable to the tumor associated antigen selected from the group consisting of BFP, AFP, TAG and CEA or the antibody thereagainst constituting the bound product, or reacting the immune complex-insolubilized substance bound product with a predetermined amount of labelled anti-complement, anti-antibody or protein A. (3) separating the resulting reaction product from the unreacted labelled substance or from the unreacted labelled anti-complement, anti-antibody or protein A. and (4) determining the specific activity of the reaction product or the separated unreacted reactant. The method is useful for diagnosis of cancers.
