    Abstract: PURPOSE:To provide an apparatus to successively transfer a specimen to a step for supporting a protein or peptide on a specimen carrier having a specific structure, a step for drying the specimen, a step for hydrolyzing by solid-gas reaction, a step for extracting the hydrolyzed specimen and a step for introducing the extracted liquid to an amino acid analyzer. CONSTITUTION:A protein or peptide is supported on a glass filter 7 of a specimen carrier 9 composed of an inner glass cylinder 6 holding a glass filter 7 for supporting a specimen and an outer metal cylinder 8 holding said inner cylinder 6 and the specimen is dried in a vacuum box 10. The specimen carrier 9 is transferred to a hydrolysis stage and hydrolyzed hy a solidliquid reaction under the supply of an acid-mixed steam to the specimen under high temperature and pressure condition. The hydrolyzed specimen is extracted with dilute hydrochloric acid and the obtained solution of the specimen is introduced into a sample coil 28 in a six-way valve 27 used as an introducing device for an amino acid analyzer. Each step is continuously performed without using manual operation to suppress the contamination, exclude the error caused by individual difference and enable more accurate analysis of amino acid composition.

    Abstract: PURPOSE:To obtain a device which supports protein or peptide on a specimen holder of specific structure, dries, hydrolyzes by solid-vapor phase reaction, extracts the hydrolyzed specimen, feeds the extracted solution to an amino acid analyzer and successively transports each specimen at every stage. CONSTITUTION:A glass adhesive is impregnated into an outer edge part of a glass filter 6 and solidified to give a specimen holder 8 having formed a holding part 7. Protein or peptide is supported on a glass filter 6 of the specimen holder and the specimen is dried in a vacuum box 10. The specimen holder 8 is sent to a hydrolyzing stage, acid mixed steam is fed to the specimen under pressure by heating and hydrolysis reaction is carried out by solid phase-liquid phase reaction. Then the hydrolyzed specimen is extracted with dilute hydrochloric acid and a solution having dissolved the specimen is sent to a sample coil 26 in a six-way valve 27, a feeder of amino acid analyzer. By continuously carrying out each operation without using manual operation, contamination can be reduced and difference in individual can be eliminated so that accurate amino acid composition is analyzed.

    Abstract: PURPOSE:To obtain the title device capable of carrying out accurate amino acid analysis by reducing contamination and eliminating difference in individual by making transportation of specimen supported on a specimen carrier, hydrolysis by solid phase-gas phase reaction, extraction and injection to amino acid analyzer into an on-line system. CONSTITUTION:A glass filter 7 of a specimen holder 8 is impregnated with a solution of specimen by using a microsyringe 9, stored in a reduced-pressure box 10, a solvent in the solution of specimen is removed to dry the specimen. The specimen holder 8 is fixed to a vaporizing chamber 15 and a glass tube 16, heated by a heater 14, an acid mixed solution 17 from which oxygen is purged by an inert gas is sent through a three-way cock 18 to the vaporizing chamber 15, the specimen is hydrolyzed with the acid made into vapor and then steam is sent to the chamber to remove the residual acid mixed vapor. The specimen holder 15 is transported to a stage carrying out extraction and transportation of solution, a diluted hydrochloric acid 24 is sent to extract the specimen and a liquid having dissolved the specimen is poured into a sample coil 28 in a six-way valve 27, an injector of amino acid analyzer.

    Abstract: PURPOSE: To obtain an improved hydrolyzed protein excellent in flavor when used in soup, etc., and suitable for use as a flavoring substance by decomposing and removing monochloropropanediol, etc., from a hydrolyzed protein by treatment at a specified pH and a specified temp. CONSTITUTION: Hydrochloric acid is added to a vegetable protein such as casein and they are boiled and neutralized preferably to pH 4.0-6.0 to form a hydrolyzate. An aq. soln. of this hydrolyzate is adjusted to pH 5.5-8.0 and hydrolysis is carried out at 20-180 deg.C for 5min to 10 days. Monochloropropanediol and dichloropropanol are then removed by steam distillation or other method to obtain the objective improved hydrolyzed protein.

    Abstract: PURPOSE:To obtain an amino acid having high purity and useful over a wide range of fields such as food, feed, medicine, cosmetic and industrial chemical, by contacting an aqueous solution of amino acid with a resin having a chelating functional group coordinated with a metal ion and eluting the adsorbed amino acid. CONSTITUTION:An aqueous solution of amino acid is brought into contact with a resin having chelating functional group of aminocarboxylic acid-type or aminophosphonic acid-type containing a coordinated metal ion. The contacting process is carried out at 0-100 deg.C e.g. by passing the aqueous solution of amino acid through a column packed with the resin to effect the absorption of amino acid to said resin. The adsorbed amino acid is eluted to separate a purified amino acid. After eluting and separating the amino acid, the used resin can be reused for the separation and purification of the amino acid solution.

    Abstract: PURPOSE:To extremely easily obtain the titled compound useful as food and drug, etc., in high purity, by contacting a polymer having amino group coordinated with metal ion with an aqueous solution of an amino acid and eluting the amino acid. CONSTITUTION:An amino acid is purified and separated by contacting an aqueous solution of amino acid with a polymer having amino group coordinated with a metal ion (e.g. transition metal ion, especially ion of Fe, Co, Ni, Cu, Zn, etc.) and eluting the amino acid from the polymer. Especially excellent effect of the process can be attained in the separation and purification of leucine containing isoleucine, methionine, etc., as impurities and produced by the alkali neutralization and precipitation of an acid-hydrolyzed casein, keratin, hemoglobin, etc. The reaction is carried out by adjusting the pH of the amino acid aqueous solution to about 2-12.
