A process for producing an .alpha.-L-aspartyl-L-phenylalanine lower alkyl ester or its hydrochloride which comprises (a) treating an L-N-(1-methyl-2-lower aliphatic acylvinyl or 1-methyl-2-lower alkoxycarbonylvinyl)-aspartic acid or an alkali metal salt thereof with a dehydrating agent in an organic solvent containing an acid, (b) reacting the solution resulting from such treatment with an L-phenylalanine lower alkyl ester and (c) treating the resulting reaction mixture with water or an aqueous acid solution to remove the 1-methyl-2-lower aliphatic acylvinyl or 1-methyl-2-lower alkoxycarbonylvinyl group from the L-N-(1-methyl-2-lower aliphatic acylvinyl or 1-methyl-2-lower alkoxycarbonylvinyl)-aspartyl-L-phenylalanine lower alkyl ester is disclosed, along with L-N-(1-methyl-2-acetylvinyl)-aspartic acid anhydride.

A compound of D-phenylalanine derivative for hypoglycemic use, represented by the general formula ##STR1## R.sup.1 is selected from hydrogen, alkyl of 1 to 5 carbon atoms, aryl of 6 to 12 carbon atoms, aralkyl of 6 to 12 carbon atoms, ##STR2## --CH.sub.2 CO.sub.2 R.sup.3, --CH(CH.sub.3)--OCO--R.sup.3, and --CH.sub.2 --OCO--C(CH.sub.3).sub.3 ; R.sup.2 is selected from groups comprising aryl of 6 to 12 carbon atoms, a hetero six-membered ring, a hetero five-membered ring, cycloalkyl, or cycloalkenyl, any of which groups may have one or more substituents; and R.sup.3 is selected from hydrogen and alkyl of 1 to 5 carbon atoms; the salts thereof, and precursors which can be converted thereto in the human or animal body.Some of the compounds are novel per se.

Amino acids having masked amino groups react with amino acids having masked carboxyl groups to form peptide derivatives in good yields if the reaction medium contains an isonitrile. Isonitriles also are effective in causing condensation of amino acids having masked amino groups with compounds having active hydroxyl groups to the corresponding amino acid esters. The yields of the first-mentioned reaction are enhanced if the reaction mixture additionally contains one of the compounds having active hydroxyl groups. Both the peptide derivatives and the amino acid esters of the compounds having active hydroxyl groups are useful for peptide synthesis.
