Abstract: Provided is a modified nucleic acid that inhibits microRNA, and a use thereof. The modified nucleic acid that inhibits microRNA, according to the present invention, can strongly bond with microRNA by consecutively arranging at least two micro RNA target sites comprising a complementarily-bonding nucleotide sequence of a minimum of six on a micro RNA seed region, and, as a result of the insertion of an abasic spacer between the micro RNA target sites, not only effectively inhibits the micro RNA, but also has the benefit of the sequence inserted in the modified nucleic acid preventing inadvertent bonding with another micro RNA, and not only canonical target but also non-canonical target recognition of micro RNA such as a nucleation bulge site and G:A or G:U wobble arrangement sites can be used, and it is possible to selectively inhibit biological functions taking place due to non-canonical target recognition of micro RNA.

Abstract: The present invention relates to RNA interference-inducing nucleic acid that inhibits noncanonical target genes of micro RNA, in which part of the sequence of a specific micro RNA has been modified, and by using the RNA interference-inducing nucleic acid of the present invention, the biological function micro RNA exhibits by inhibiting noncanonical target genes is effectively increased or there is the benefit of selectively exhibiting only one of the biological functions of conventional micro RNA, i.e., the function of inhibiting noncanonical target genes, and the interference-inducing nucleic acid of the present invention enables cell cycling, differentiation, dedifferentiation, formation, movement, splitting, proliferation or death adjustment, and it is expected that the invention can be used in various fields such as drugs and cosmetics.

Abstract: In the present invention, it has been confirmed that, when an RNA interference-inducing nucleic acid including at least one 8-oxoguanine (o8G) in 1st to 9th nucleotides from the 5′-end of at least one single strand of a double strand of a nucleic acid, and a modified nucleic acid that specifically binds to microRNA and in which at least one guanine (G) from among the 1st to 9th nucleotides from the 5′-end are modified with 8-oxoguanine (o8G), are produced and administered to cells or mice, various pathophysiological phenomena are induced.
In addition, the positions where G>T modifications occur have been identified in cDNA produced through the reverse transcription of microRNA in which guanine (G) is oxidatively modified with 8-oxoguanine (o8G) by oxidative stress in a seed region of microRNA, to confirm the positions where oxidative modification to 8-oxoguanine has occurred.
