    摘要: A method of purifying human growth hormone (hGH) from deamidated hGH, oxidized hGH or both. The method comprises the steps of forming a metal cation complexed-hGH composition enriched in native hGH under conditions wherein none or less than all of the deamidated hGH, oxidized hGH or both complex with a metal cation and, isolating the metal cation-complexed hGH composition from the deamidated hGH, oxidized hGH or combination thereof. The method can further comprise the step of releasing purified hGH from the complex. Alternatively, the metal cation-complexed hGH can be encapsulated into a biocompatible polymer for sustained release of hGH. The purification method can be performed one or more times using the hGH released from the metal cation-complexed hGH composition, depending on the purity of hGH desired.

    摘要: Described herein is a method of purifying a target molecule from an aqueous biological composition, the method comprising: (a) contacting a cationic polymer and the aqueous biological composition to form a mixture, the mixture comprising a bio-polymer complex and the target non-binding molecule in a liquid, wherein the bio-polymer complex has an average particle diameter of at least 50 micrometers; (b) providing a filtering unit comprising (i) a housing having an inlet and an outlet, (ii) a porous, continuous filter medium which is fluidly connected to the inlet and the outlet, and (iii) a collection region upstream from the porous, continuous filter medium; (c) adding the mixture to the inlet; and (d) allowing the mixture to separate in the filtering unit, whereby the bio-polymer complex collects in the collection region and the target non-binding molecule passes through the filter medium, and wherein the majority of flow of the liquid through the porous, continuous filter medium is not substantially parallel with the direction of gravity.

    摘要: The invention provides methods for a purifying protein of interest from a mixture comprising the protein of interest and one or more contaminants, including host cell DNA and proteins, by precipitation of the contaminants with caprylic acid. Such methods are particularly useful for purifying antibodies from cell cultures. Moreover, mixtures that have been depleted of contaminants using the methods of the invention can be used directly in downstream chromatography applications (e.g., ion exchange chromatography) without any further purification. These methods lead to manufacturing processes with a minimum number of unit operations and reduce the resource requirements, and thus positively influence the cost of goods for therapeutic protein production.

    摘要: A method is provided for purifying a protein comprising the steps of providing a heme tag with an open coordination site and tagging the recombinant protein of interest with the heme tag. A resin framework is used, wherein a base that binds to heme is immobilized to the resin, and the open coordination site of the heme tag is capable of reversibly binding to the base immobilized to the resin. The tagged protein is reversibly bound to the resin, then eluted from the resin and quantified. The method enables the tagged protein to be tracked during protein expression or purification, and can be used to identify secretion of a protein to the periplasm or to tag proteins in the cytoplasm.
