    Abstract: The present invention provides a system and a method of producing large capacity cloning vectors, preferably BACs carrying an intact viral genome, by direct cloning of virion DNA or of circular or head-to-tail concatemeric DNA replication-intermediates. These replication-intermediates are digested with a chosen restriction enzyme that cleaves once in the genome and the resultant linear DNA corresponding to the intact genome is cloned into a BAC vector, engineered to contain the chosen restriction enzyme. In one embodiment the present invention concerns a system and a method for producing BAC comprising the entire HHV-6A genome. The resultant BAC-HHV-6A clones can be used in gene therapy, in vaccination, as well as in the study of viral functions, and their association with disease.
