The present invention provides a D-psicose-3-epimerase mutant for efficient and effective production of D-allulose (also known as D-psicose) from D- fructose. The invention represents an advancement in the field of enzyme engineering. The mutant enzyme has increased thermo-stability, operational stability and is functional at wide range of pH as compared to enzymes existing in the prior art and can be used for obtaining high yield of D-allulose with minimum downstream processing cost. Further, the invention relates to nucleic acid encoding the said protein, expression cassettes, vectors, recombinant host cells for preparing the mutant enzyme and processes for preparing the mutant enzyme and processes for production of D-allulose from D-fructose using the mutant enzyme.

The present invention discloses a low calorie and low glycemic index (GI), and sustained energy release brown sugar substitute composition. The brown sugar substitute provides a suprising effect regarding cognitive performance, such as mood, of a subject compared to individual constituents. The brown sugar substitute comprises first sugar selected from a group comprising isomaltulose, trehalulose, D-allulose, isomaltulose- silicate, trehalulose-silicate and D-allulose-silicate; molasses or second sugar syrup; wherein the second sugar syrup is selected from a group comprising brown isomaltulose syrup, brown trehalulose syrup and brown allulose syrup. The present invention also disclosed a process for preparing the brown sugar substitute.

The present disclosure relates to a process for separating D-Allulose from an aqueous feed mixture comprising D-Allulose and at least one other ketose, particularly, fructose. The process comprises: adding calcium chloride to the solution comprising D-Allulose and fructose to get a mixture; drying the mixture obtained in the above step; adding aqueous ethanol to the dried mixture to precipitate the fructose as a hydrated addition compound of said calcium chloride; filtering the precipitate formed and recovering the fructose from the precipitate; collecting the filtrate comprising calcium addition D-Allulose and recovering the D-Allulose from the filtrate.
