Abstract: A chemical approach is provided to profile uptake of compounds in cells, alone or in combination with other analytical techniques. The disclosure is particularly well suited for detecting cellular uptake of metabolites such as fatty acids. The disclosure can be carried out, applied in solution or on a surface, in bulk or on single cells, and is compatible with one or more additional techniques such as protein analysis. The disclosure is exemplified by probing fatty acid influx alone and in combination with proteomics analysis on a single-cell barcode chip to identify a combination therapy for inhibiting fatty acid metabolism and treating cancer.

Abstract: This disclosure provides electrochemically-cleavable linkers with cleavage potentials that are less than the redox potential of the solvent in which the linkers are used. In some applications, the solvent may be water or an aqueous buffer solution. The linkers may be used to link a nucleotide to a bound group. The linkers include a cleavable group which may be one of a methoxybenzyl alcohol, an ester, a propargyl thioether, or a trichloroethyl ether. The linkers may be cleaved in solvent by generating an electrode potential that is less than the redox potential of the solvent. In some implementations, an electrode array may be used to generate localized electrode potentials which selectively cleave linkers bound to the activated electrode. Uses for the linkers include attachment of blocking groups to nucleotides in enzymatic oligonucleotide synthesis.

Abstract: Methods for separating, in a continuous, multizone fluid medium, cells, particles, or other molecules of interest (MOI) from associated or contaminating unwanted materials not of interest (MNOI). The invention involves forced movement of MOI into certain zones having properties which deter the entry of unwanted materials. Differential movement of MOI and MNOI occurs by active counterforces that move MNOI but not MOI. MOI are tagged with magnetic particles and moved with a magnetic field through a fluid, or zones, of higher specific gravity that prevents, by flotation counterforce, unwanted less dense materials from entering. Surfaces specifically coated with reactants are reactive with the MOI in the tagged magnetic particle complex and of buoyant or other forces are used to remove any unbound material from the surface before reading. Readable labels, in addition to the magnetic particle tagged complex itself, such as enzymes, fluorophors, chemiluminescent materials, radioactive isotopes, chromogenic and fluorogenc substrates and other labels may be used. In most embodiments, materials of interest are delivered to a special final zone for reading or harvesting. The invention applies to many assays, diagnostic tests, separative procedures and chemical syntheses.
