A flow cytometer (10) includes a plurality of detectors (28, 30, 32, 34, 36, 38) for providing voltage pulse signals over a four decade range as particles pass through an illuminated detection station (20). Each voltage pulse signal is processed and provided to a first sample and hold (84, 138) for storage. Thereafter, the value in the first sample and hold is amplified by one or thirty-two, depending upon its magnitude, and stored in a second sample and hold (84, 146) for subsequent provision to a sixteen bit analog to digital convertor (ADC) (66), one signal at a time. The first sample and hold is then free to store a new value while the old value is awaiting provision to the ADC. The ADC converts the signal received from the second sample and hold to a fifteen bit digital signal and uses the sixteenth bit to manifest whether the signal stored in the second sample and hold was amplified by thirty-two.

A method and apparatus (320, 340, 360) for automatically and rapidly retrieving, counting and/or analyzing at least one selected population of cells or formed bodies, such as a white blood cell population and at least one subset thereof of a whole blood sample (322, 342) or portion thereof. A volume of a biological medium containing the white blood cells is prepared and at least one reactant (326, 346) specific or preferential at least to some selected biological cells is introduced thereto and rapidly mixed (384) for a short period of time. The opacity and/or volume parameter of the cells can be modified and the mixture is then counted and analyzed (332, 352, 416, 422) in one or more steps to obtain the desired white blood cell population and subset analysis. The biological sample can be a whole blood sample and the reactant can include or be a lyse or a monoclonal antibody bound to microspheres, which will bind to specific ones of the cells or a combination of lyse and microspheres with antibody bound thereto. The microspheres can be magnetic and the bound cells can be magnetically removed for retrieving and analyzing the remaining blood cell population.

A method and reagent system for the rapid isolation, identification, and/or analysis of leukocytes from a whole blood sample. The reagent system includes a lytic reagent solution comprising an aqueous solution of a water soluble compound which at least partially dissociates in aqueous solution to release a proton and a counterion and which, upon contact with a blood sample, acidifies the sample to a pH in the range of from about 2.6 to 4.0. The reagent system also includes a quenching reagent solution comprising an aqueous alkaline salt solution which effectively and rapidly arrests the chemical action of the lytic reagent solution on the sample and restores the native physiological environment of the sample. The method comprises using the reagent system to selectively lyse the red blood cells in a blood sample and modify the leukocytes to enable their differentiation into five sub-populations.

A method for preparing phycobiliprotein/amine-reactive dye conjugates is disclosed in which the conjugates so prepared overcome the energy transfer/fluorescent quenching dilemma encountered in the use of prior art conjugates. A phycobiliprotein, for example, phycoerythrin or allophycocyanin, is conjugated with an amine-reactive dye, for example, Texas Red or carboxyfluorescein succinimidyl ester, in the presence of a selective salt which causes a hydrophobic intramolecular rearrangement of the phycobiliprotein thereby exposing more hydrophobic sites for binding to the amine-reactive dye. The conjugates prepared according to the invention are useful in multiple color fluorescence assays without requiring the use of multiple exciting sources.

L'aminodextrane agit aussi bien comme réducteur pour réduire les ions métal en particules de métal(O) que comme agent protecteur qui recouvre les particules de métal(O) ainsi formées. Après stabilisation du revêtement d'aminodextrane à l'aide d'un agent réticulant, les particules revêtues peuvent être utilisées pour lier des protéines de manière covalente. Les particules colloïdales contenant des protéines résultantes peuvent être utilisées comme marqueurs en microscopie optique et électronique, dans des essais immunologiques et biologiques, et éventuellement comme agents thérapeutiques.

The reaction involves selected viable biological cells which are prepared in a small sample volume and rapidly mixed with microspheres having antibody specific at least to specific ones of the cells bound thereto. The microspheres can be magnetic and the bound cells can be magnetically removed to analyze the remaining blood cell populations. The microspheres can be introduced sequentially or simultaneously.
