The purpose of the present invention is to provide a technique to duplicate and translocate any large region in a chromosome of a fungus belonging to Aspergillus across a wide range, so that it will possible to stably and systematically acquire an Aspergillus strain having a novel trait, which was un-acquirable by conventional techniques. The present invention relates to a transformant of the fungus belonging to Aspergillus wherein a transformation marker gene with deficiency of a terminal part at the 5' or 3' end of its coding region is integrated into an outside of a target region in a chromosome of the fungus subject to duplicated translocation, and a transformation marker gene with deficiency of a terminal part at the 3' or 5' end of its coding region is integrated into an outside of a region in another chromosome of the fungus to be replaced with the target region.

The present invention provides heat-resistant flavin-bound glucose dehydrogenases having high substrate specificity for D-glucose, an efficient production method of the same, and a transformant used for the same. A flavin-bound glucose dehydrogenase gene encoding a flavin-bound glucose dehydrogenase derived from Mucor is introduced into yeast, Zygosaccharomyces, to obtain a transformant. Subsequently, the yeast transformant is cultured to obtain a flavin-bound glucose dehydrogenase from the culture. A heat-resistant flavin-bound glucose dehydrogenase that is less susceptible to the effects of dissolved oxygen and allows accurate measurement of glucose even in the presence of sugar compounds other than glucose in a sample can be efficiently produced.

Such amadoriase comprises substitution of one or more amino acid residues at positions corresponding to amino acids selected from the group consisting of position 98, position 259, position 154, position 125, position 261, position 263, position 106, position 103, position 355, position 96, position 66, position 67, position 70, position 100, position 110, position 113, position 114, and position 156 in the amadoriase derived from the genus Coniochaeta. This invention enables accurate measurement of α-fructosyl valyl histidine derived from the β-chain amino terminus in glycated hemoglobin in the presence of ε-fructosyl lysine.

There are provided flavin-binding glucose dehydrogenases with high substrate specificity for D-glucose. Novel flavin-binding glucose dehydrogenases which are derived from microorganisms belonging to the genus Mucor. They have low reactivity for maltose, D-galactose and D-xylose compared to their reactivity for D-glucose, and are therefore relatively unaffected by these saccharide compounds. They are also relatively unaffected by dissolved oxygen, and allow accurate measurement of glucose amounts even in the presence of saccharide compounds other than glucose in samples.
