An imaging apparatus with an autofocus mechanism for obtaining focused images. The apparatus comprises: an objective lens, a focus controller (16, 20) for altering a distance between the objective lens and a sample, an object finder (16) for finding objects of interest within the sample, and a light intensity measurement unit (14) which measures light intensity levels of the the identified objects of interest. The focus control alters the sample-objective distance to maximize the light intensity levels being measured, to thereby arrive at a focus position. Objects of interest may be identified by filtering out large objects and optionally by masking out background regions. The apparatus is useful for microscopy and for fluorescent imaging in which light levels are low and noise is high.

Methods and apparatus are provided for computing focus information prior to scanning microscope slides with a line scan camera. The methods include a point-focus procedure that works by moving the slide to the desired measurement location, moving the objective lens through a predefined set of height values, acquiring imagery data at each height, and determining the height of maximum contrast. The methods also include a ribbon-focus procedure whereby imagery data are acquired continuously, while the slide and objective lens are in motion. Both methods may be applied with either a static or a dynamic implementation.

A method and apparatus for automated cell analysis of biological specimens automatically scans at a low magnification to acquire images (288) which are analyzed to determine candidate cell objects of interest. The low magnification images are converted from a first color space to a second color space (290). The color space converted image is then low pass filtered (292) and compared to a threshold (294) to remove artifacts and background objects from the candidate object of interest pixels of the color converted image. The candidate object of interest pixels are morphologically processed (296) to group candidate object of interest pixels together into groups which are compared to blob parameters (298) to identify candidate objects of interest which correspond to cells or other structures relevant to medical diagnosis of the biological specimen. The location coordinates of the objects of interest are stored and additional images of the candidate cell objects are acquired at high magnification. The high magnification images are analyzed in the same manner as the low magnification images to confirm the candidate objects of interest which are objects of interest. A high magnification image of each confirmed object of interest is stored for later review and evaluation by a pathologist.

The invention relates to a method and apparatus for focusing structured image such as a laser spot. A method for determining the degree of focus of such a structured image is disclosed whereby: an image is provided at a photosensitive array formed from a plurality of elements; the position of the image is determined; for an element of the array a weighting is assigned according to the position of that element relative to the position of the image; the intensity of the image at the element is determined; a value is produced for the element dependent on its weighting and the intensity of the image thereat. In addition the values of the plurality of elements are summed and one overall focus value is found which is indicative of the degree of focus of the image. This value is used to search for and maintain best focus and methods for achieving this are disclosed also. Apparatus is shown in which a microprocessor and a controller are used to perform the methods disclosed.
