    摘要: The present invention provides a preparation and application device useful for mixing and dispensing a plurality of fluids, wherein the fluids must be mixed immediately prior to use. In one embodiment, the device includes two fluid delivery systems connected to a manifold having two separate fluid channels. Each fluid channel conducts fluid to a mixing chamber, from which, following mixing, the mixture of fluids is dispensed. The manifold additionally includes a gas channel having an outlet connected to one of the fluid channels, which can be used to supply a pressurized gas to the mixing chamber to expel the mixture from the chamber, reducing the likelihood that the mixture can clog the device. The invention also provides a method for preparing and dispensing a mixture of two fluids.

    摘要: Electrophoretic chambers having at least a region of surface modification, and methods for their fabrication, are provided. In some embodiments the subject chambers include in the region of surface modification an anchoring polymeric layer interpenetrating the surface of the chamber and an electrophoretic polymeric layer copolymerized with the anchoring polymeric layer. The subject chambers are prepared by sequentially contacting the chamber surface with a first monomer capable of interpenetrating the surface and a second monomer capable of copolymerization with the first monomer, followed by copolymerization of the first and second monomers. In other embodiments an electrophoretic polymeric layer is noncovalently bound on the surface of a rigid polymeric base material without the aid of a separate anchoring polymeric layer. The subject devices find use in any of a variety of electrophoretic applications in which entities are moved through a medium under the influence of an applied electric field.

    摘要: Libraries of novel backbone-cyclized peptide analogs are formed by means of bridging groups attached via the alpha nitrogens of amino acid derivatives to provide novel non-peptidic linkages. Novel building units used in the synthesis of these backbone-cyclized peptide analogs are Nα (φ-functionalized) amino acids constructed to include a spacer and a terminal functional group. One or more of these Nα (φ-functionalized) amino acids are incorporated into a library of peptide sequences, preferably during solid phase peptide synthesis. The reactive terminal functional groups are protected by specific protecting groups that can be selectively removed to effect either backbone-to-backbone or backbone-to-side chain cyclizations. The invention is exemplified by libraries of backbone-cyclized bradykinin analogs, somatostatin analogs, BPI analogs and Substance P analogs having biological activity. Further embodiments of the invention are Interleukin-6 receptor derived peptides having ring structures involving backbone cyclization.

    摘要: Novel acid-stable and thermo-stable enzymes having α-1,4 hydrolytic activity and a α-1,6 hydrolytic activity which are derived from strains of the genus Sulfolobus. These enzymes are capable of expressing high levels of α-1,4 hydrolytic activity, including the maximum α-1,4 hydrolytic activity thereof, at highly acidic pHs of between about 2.5 and about 4.5. These α-amylases are further capable of expressing high levels of α-1,4 hydrolytic activity, including the maximum α-1,4 hydrolytic activity thereof, at high temperatures of between about 90 °C and about 120 °C. Particularly disclosed herein are such enzymes which are derived from strains of the species S. acidocaldarius and, in particular, Sulfolobus acidocaldarius DSM 639. Modified starch degradation (liquefaction and saccharification) processes using these novel enzymes are also disclosed herein.
