    摘要: There is provided a method of regulating a eukaryotically active gene, comprising transforming a cell with a transformation cassette expressing a modulator gene product regulating the eukaryotically gene or its product and a further gene product regulating said modulator gene or its product, the promoters of two of said gene, modulator gene and further genes being selected from inducible promoters and developmental promoters for the same or complementary tissues. The lethal gene expressing barnase, a ribonuclease of B. amiloliquefaciens, is placed under the control of a tissue specific promoter, such as those derived from PrMADS1, 2 or 3 of Pinus radiata or EGM1, 2 or 3 of Eucalyptus grandis. The same tissue specific promoter is used to express Laclq gene, a repressor for barnase (barstar) being promoted by a modified 35s RNA CaMV promoter including the lac operon. The cassette is used to transform plant cells for regeneration into plants expressing the barnase in the target tissues with improved specificity and reduced promoter leakage.

    摘要: A method of calibrating a ribbon winding mechanism (10) of a printing apparatus which is adapted to perform a printing operation in which at a printing station (20) a print head (11) is moved relative to a first length of the ribbon (13) to tranfer print medium from the first length of the ribbon (13) onto a substrate and the ribbon (13) is subsequently advanced a predetermined amount to present a second length of ribbon (13) at the printing station (20) for use in a further printing operation, the winding mechanism (10) comprising a supply spool (11) and a take-up spool (14) and there being a ribbon path between the supply (11) and take-up spools (14) through the printing station (20), and a motive means (M) to cause the ribbon (13) to be moved along the ribbon feed path and wound onto the take-up spool (14), and a sensing means (22) to sense movement of the ribbon (13) as the take-up spool (14) is rotated and to provide a signal to a control means (25) in response, the method being characterised in that prior to performing the printing operation, the take-up spool (14) is rotated a predetermined amount whilst the sensing means (22) senses the resultant ribbon (13) movement along the ribbon feed path and the control means (25) determines from the signal received from the sensing means (22) the amount to rotate the take-up spool (14) in the subsequent printing operation to advance the ribbon (13) the predetermined amount.
