An anti-feline herpesvirus-1 recombinant antibody which is efficacious in treating, preventing and diagnosing feline herpesvirus-1 (FHV-1) infection, and a gene fragment useful for the preparation of the antibody. A gene fragment coding for the antibody V region bearing an FHV-1 specific avidity of a mouse monoclonal antibody having an excellent activity of neutralizing FHV-1 is obtained by constructing cells which produce the antibody. A chimeric anti-FHV-1 recombinant antibody is prepared from the above gene fragment and another gene fragment coding for a feline antibody C region.

It is intended to provide a novel preventive/remedy for diseases with immune abnormality which can effectively act as a super-antigen without being neutralized by a neutralizing antibody to Staphylococcus enterotoxin B (SEB) known as a super-antigen. Namely, an SEB modification having a lowered reactivity with a neutralizing antibody to SEB (anti-SEB antibody) and a preventive/remedy for diseases with immune abnormality which contains this modification as the active ingredient. This SEB modification can be obtained via amino acid substitution in the amino acid sequence of SEB, in particular the amino acid sequence at the epitope part recognized by the anti-SEB antibody by using evolutionary molecular engineering techniques.

The invention relates to a gene fragment which codes for the variable regions of an antibody having an activity of neutralizing human immunodeficiency virus (HIV), a novel recombinant anti-HIV antibody expressed by using the above gene, and a process for preparing the same. A mouse-human chimeric antibody and a modified mouse-human antibody, having an activity of neutralizing HIV, are obtained by determining a specific nucleic acid sequence of a gene fragment which codes for the variable regions of the H and L chains of an antibody having an activity of neutralizing HIV and artificially fusing a DNA synthesized on the basis of the determined sequence with a gene which codes for human immunoglobulin. The novel anti-HIV antibody is useful for treating and preventing AIDS.

The invention provides a clinically useful monoclonal antibody which can recognize the conserved region of the V3-PND region of a glycoprotein antigen (gp120) having a molecular weight of about 120,000 Da and contained in the coat of a human immunodefiency virus (HIV) and which can neutralize a wide variety of HIV variants, or a fragment of the above antibody, and chimera and humanized antibodies derived from the above antibody. It is possible to prepare a monoclonal antibody having the activity of neutralizing various HIV variants by using as the immunogen several types of peptides having a PND tip region containing a GPGR sequence having a high conservability in the PND region of the gp120 of an HIV. It is possible to obtain a chimera antibody or an altered antibody having an anti-HIV neutralization activity effective for clinical application by transplanting a fragment of a gene coding for the variable region of the above monoclonal antibody or the complementarity-determining region of that region to a human antibody gene.
