摘要: In order to identify an antigen to 2D7 antibody, a 2D7 antigen is cloned. As a result, it is indicated that the 2D7 antigen is an HLA class I molecule. Based on this finding, it is discussed whether the 2D7 antibody has an activity of inducing cell death or not. As a result, nuclear fragmentation is observed by further crosslinking the 2D7 antibody with another antibody, thereby indicating that cell death is induced. It is also clarified that a diabody of the 2D7 antibody has an extremely strong cell death-inducing activity even though no other antibody is added. These results point out that a degradation product of an antibody recognizing HLA is usable as a cell death-inducing agent.

摘要: Cloning of antigen 2D7 has been carried out to identify an antigen relevant to antibody 2D7. As a result, it has been found that the antibody 2D7 recognizes HLA class IA. Further, whether or not the antibody 2D7 has cell death inducing activity has been studied. As a result, it has been found that nuclear fragmentation is observed by crosslinking of the antibody 2D7 with another antibody to thereby bring about induction of cell death. Still further, it has been found that a diabody of antibody 2D7 exhibits very strong cell death inducing activity even when any other antibody is not added. These results suggest that an antibody with lowered molecular weight among antibodies capable of recognizing HLA can be utilized as a cell death inducer.

摘要: A test is performed on the effects of an interferon on the expression of HLA-A and the cytotoxicity caused by an anti-HLA class I antibody in a cell, using a 2D7-DB antibody as the anti-HLA class I antibody and an IM-9 cell strain as the cell to be tested. As a result, it is demonstrated that the expression of HLA-A can be up-regulated by IFNα and IFNϜ. Further, a WST-8 assay performed using an interferon and an anti-HLA class I antibody in combination to examine the cell viability demonstrates that cell death activity can be increased by the combination of 2D7-DB and IFNα or IFNϜ.
